【摘要】: 研究表明應(yīng)激能夠影響動(dòng)物和人體的免疫功能,機(jī)體免疫系統(tǒng)功能的紊亂是感染性疾病、腫瘤等發(fā)生或惡化的直接原因,故闡明應(yīng)激對(duì)免疫系統(tǒng)損傷的作用機(jī)制具有重要意義。動(dòng)物模型具有資源相對(duì)易獲得、操作性強(qiáng)、涉及倫理道德問題相對(duì)較少等優(yōu)點(diǎn),因此建立理想的應(yīng)激動(dòng)物模型是研究得以深入的關(guān)鍵。我們應(yīng)用束縛應(yīng)激和熱應(yīng)激建立了兩種小鼠應(yīng)激動(dòng)物模型,然后使小鼠染菌,通過比較應(yīng)激狀態(tài)下小鼠與正常小鼠的死亡率來研究兩種應(yīng)激對(duì)小鼠免疫防御功能的影響,并試圖從遺傳背景角度揭示基因多態(tài)性與應(yīng)激狀態(tài)下免疫應(yīng)答強(qiáng)度的關(guān)系;通過應(yīng)激所導(dǎo)致的機(jī)體免疫器官損傷、血清中蛋白質(zhì)的變化及全血中金屬元素含量的變化幾個(gè)方面來揭示應(yīng)激對(duì)免疫防御功能影響的相關(guān)機(jī)制。 方法 應(yīng)用束縛應(yīng)激和熱應(yīng)激建立了兩種小鼠應(yīng)激動(dòng)物模型,然后使小鼠染菌,研究這兩種應(yīng)激對(duì)小鼠免疫防御功能影響,并通過以下幾個(gè)方面來研究應(yīng)激對(duì)免疫功能損傷的可能機(jī)制。 1.研究應(yīng)激對(duì)小鼠脾臟重量指數(shù)的影響,觀察接受應(yīng)激的小鼠脾臟的病理變化,闡述免疫防御功能降低的構(gòu)效關(guān)系。 2.以已經(jīng)建立的動(dòng)物模型為研究對(duì)象,采用PCR技術(shù)檢測(cè)以MudoEb5和MudoEb7為代表的小鼠H2-Eb位點(diǎn)等位基因的多態(tài)性,從遺傳多樣性的角度揭示基因多態(tài)性與應(yīng)激狀態(tài)下免疫應(yīng)答強(qiáng)度的關(guān)系。 3.采用SELDI-TOF-MS技術(shù),運(yùn)用弱陽離子交換芯片(WCX2)對(duì)小鼠血清蛋白質(zhì)指紋圖譜進(jìn)行研究,探索不同類型的應(yīng)激對(duì)免疫功能影響的特點(diǎn)及物質(zhì)基礎(chǔ)。
[Abstract]:Studies have shown that stress can affect the immune function of animals and human beings, and the disorder of immune system is the direct cause of infectious diseases, tumors and so on. Therefore, it is of great significance to clarify the mechanism of stress on immune system damage. Animal models have the advantages of relatively easy access to resources, strong maneuverability, and relatively few ethical and moral problems. Therefore, the establishment of ideal animal models of stress is the key to further study. We used restraint stress and heat stress to establish two kinds of animal models of mouse stress, and then made the mice infected with bacteria. By comparing the mortality of mice under stress and that of normal mice, the effects of two kinds of stress on the immune defense function of mice were studied. From the perspective of genetic background, this paper attempts to reveal the relationship between gene polymorphism and the intensity of immune response under stress. The mechanism of the effect of stress on immune defense function was revealed from several aspects, such as the damage of immune organ, the change of protein in serum and the change of metal element content in whole blood. Method Two kinds of animal models of mouse stress were established by restraint stress and heat stress, and then the mice were infected with bacteria to study the effects of these two stresses on the immune defense function of mice. And through the following aspects to study the possible mechanism of stress damage to immune function. 1. To study the effect of stress on spleen weight index of mice, observe the pathological changes of spleen of mice receiving stress, and elucidate the structure-activity relationship of the decrease of immune defense function. 2. The alleles of H2-Eb locus in mice represented by MudoEb5 and MudoEb7 were detected by PCR technique. From the perspective of genetic diversity, the relationship between genetic polymorphism and the intensity of immune response under stress was revealed. 3. The fingerprint of serum protein in mice was studied by using SELDI-TOF-MS and WCX2 (weak cationic exchange chip). To explore the characteristics and material basis of the effects of different types of stress on immune function.
