【摘要】: 目的:研究在端腦發(fā)育中骨形態(tài)發(fā)生蛋白4(BMP-4)對(duì)同源域轉(zhuǎn)錄因子Nkx2.1表達(dá)的影響,探討Nkx2.1在膽堿能神經(jīng)元發(fā)生過程中的作用以及能否被BMP-4所誘導(dǎo)。方法:將E16大鼠胚胎端腦細(xì)胞做體外無血清原代培養(yǎng),用不同濃度的BMP-4誘導(dǎo)膽堿能神經(jīng)元的分化,采取免疫細(xì)胞化學(xué)染色實(shí)驗(yàn)觀察不同濃度BMP-4對(duì)ChAT陽性免疫反應(yīng)細(xì)胞數(shù)量的影響。在實(shí)驗(yàn)過程中采取對(duì)膽堿能神經(jīng)元分化影響最大的BMP-4濃度,加入到體外培養(yǎng)的E16大鼠胚胎端腦細(xì)胞中,進(jìn)行RT-PCR檢測(cè),以研究Nkx2.1在端腦發(fā)育中BMP-4對(duì)Nkx2.1基因表達(dá)的影響。結(jié)果:不同濃度BMP-4對(duì)膽堿能神經(jīng)元發(fā)生的影響有所不同。低濃度(10ng/ml、20ng/ml)BMP-4可使ChAT陽性細(xì)胞數(shù)量增多;高濃度(40ng/ml)BMP-4可使ChAT陽性細(xì)胞數(shù)量減少。RT-PCR結(jié)果顯示實(shí)驗(yàn)組和對(duì)照組Nkx2.1基因表達(dá)沒有明顯區(qū)別。結(jié)論:(1)在體外培養(yǎng)的E16端腦細(xì)胞中,低濃度(20ng/ml)BMP-4能促進(jìn)膽堿能前體細(xì)胞的發(fā)生和分化;(2)在體外培養(yǎng)的E16端腦細(xì)胞中,高濃度(40ng/ml)BMP-4能抑制膽堿能前體細(xì)胞的發(fā)生和分化;(3)BMP-4細(xì)胞外信號(hào)分子不能調(diào)節(jié)E16端腦細(xì)胞Nkx2.1 mRNA的表達(dá)。
[Abstract]:Objective: To study the bone morphogenetic protein 4 in telencephalon development (BMP-4) on expression of homeodomain transcription factor Nkx2.1, to investigate the effects of Nkx2.1 on cholinergic neurons during the process of action and can be induced by BMP-4. Methods: embryonic stem cells from E16 rats were cultured in vitro without serum. Primary cultures of cholinergic neurons were induced by different concentrations of BMP-4. Immunocytochemical staining was used to observe the effect of different concentrations of BMP-4 on the number of ChAT positive immunoreactive cells. During the experiment, we took the most influential BMP-4 concentration on the cholinergic neurons, and added it to the embryonic brain cells of E16 rats cultured in vitro. RT-PCR was used to detect the effect of BMP-4 on Nkx2.1 gene expression in the development of Nkx2.1. Results: the effects of different concentrations of BMP-4 on the occurrence of cholinergic neurons were different. Low concentration (10ng/ml, 20ng/ml) BMP-4 can increase the number of ChAT positive cells, and high concentration (40ng/ml) BMP-4 can reduce the number of ChAT positive cells. The results of RT-PCR showed that there was no significant difference in the expression of Nkx2.1 gene between the experimental group and the control group. Conclusion: (1) in vitro of E16 brain cells, low concentration (20ng/ml) of BMP-4 can promote the occurrence and differentiation of cholinergic progenitor cells; (2) in vitro of E16 brain cells, high concentrations of BMP-4 (40ng/ml) can inhibit the occurrence and differentiation of cholinergic progenitor cells; (3) the expression of BMP-4 extracellular signal molecules cannot regulate E16 Nkx2.1 brain cell end mRNA.
