小麥脫落酸受體基因TaPYR1介導(dǎo)植株抵御干旱逆境功能研究

脫落酸(ABA)受體PYR/PYL/RCAR通過(guò)與滲透脅迫誘導(dǎo)的ABA結(jié)合,參與植株體內(nèi)ABA介導(dǎo)的信號(hào)轉(zhuǎn)導(dǎo)過(guò)程,在調(diào)控植株干旱逆境抵御過(guò)程中具有重要的生物學(xué)功能。本研究對(duì)1個(gè)響應(yīng)干旱的小麥PYR家族成員TaPYR1分子特征、應(yīng)答干旱表達(dá)模式及其介導(dǎo)植株抵御干旱逆境的功能進(jìn)行了鑒定。主要研究結(jié)果如下:1.通過(guò)生物信息學(xué)分析方法對(duì)小麥ABA受體基因TaPYR1進(jìn)行分析,全長(zhǎng)840 bp,開(kāi)放閱讀框636 bp。TaPYR1蛋白分子量為22.621 kD,等電點(diǎn)(PI)為4.97。TaPYR1與植物種屬中部分PYR基因在氨基酸序列水平上高度同源,編碼蛋白與植物種屬部分同源蛋白在序列組成上高度相似,含有PYR家族蛋白的SRPBCC超家族保守區(qū)域(44-192 aa)。對(duì)該基因編碼蛋白的亞細(xì)胞定位試驗(yàn)結(jié)果顯示,TaPYR1經(jīng)內(nèi)質(zhì)網(wǎng)分選后定位于細(xì)胞質(zhì)膜,表明該受體蛋白在此處參與結(jié)合ABA及介導(dǎo)特定ABA信號(hào)傳遞。2.表達(dá)分析表明,TaPYR1基因在小麥根、葉中均呈明顯的干旱誘導(dǎo)表達(dá)模式,正常生長(zhǎng)條件下,小麥根、葉片內(nèi)TaPYR1轉(zhuǎn)錄本數(shù)量均較低;干旱處理下,TaPYR1表達(dá)上調(diào),表現(xiàn)在48 h處...
