鹽脅迫下景天植物的超微弱發(fā)光及其激發(fā)機制

【摘要】:本試驗以景天屬植物德景天為試驗材料進行鹽逆脅迫,并在鹽脅迫下進行活性氧自由基誘發(fā)和活性氧自由基清除處理。通過研究景天屬植物在鹽逆境脅迫下及活性氧自由基誘發(fā)和清除情況下受脅迫程度、超微弱發(fā)光(UWL)、活性氧系統(tǒng)、能量水平、光合作用等的變化,探討鹽逆境脅迫下景天植物UWL的變化及側(cè)重于活性氧方面的產(chǎn)生機制。本研究主要結(jié)果如下:1.試驗對德景天葉片在NaCl鹽脅迫下的UWL變化、生長活力、光合作用、活性氧系統(tǒng)、能量水平進行了研究。結(jié)果表明:鹽脅迫下,德景天葉片的UWL強度下降,相對含水量下降,根系活力降低,束縛水與自由水的比值上升,相對含水量與UWL之間呈高度正相關(guān),差異性顯著,束縛水與自由水比值與UWL之間呈高度負相關(guān),差異性顯著;Pn、Tr、Gs、Ci、Chla含量、Chlb含量、Chl總含量下降,光合能力減弱,UWL與Pn、Tr、Gs、Ci、Chla含量、Chlb含量、Chl總含量呈高度正相關(guān),差異性顯著;O2-產(chǎn)生速率、MDA含量、相對電導(dǎo)率上升,UWL與O2-產(chǎn)生速率、MDA含量、相對電導(dǎo)率呈高度負相關(guān),差異性顯著;ATP含量呈下降趨勢,能荷值下降,ADP和AMP含量整體上升,ATP、能荷值均與UWL呈高度正相關(guān),差異性極顯著。反映了鹽脅迫程度。初步表明光合作用,能量相關(guān)與UWL產(chǎn)生有關(guān),活性氧不是產(chǎn)生UWL的直接來源。2.試驗進一步對德景天葉片在施加過氧化氫(H2O2)和苯甲酸鈉調(diào)控鹽脅迫下的UWL變化、生長活力、光合作用、活性氧系統(tǒng)、能量水平進行了研究。結(jié)果表明:在鹽脅迫下,NaCl處理(對照)、NaCl+H2O2處理和NaCl+苯甲酸鈉處理的德景天葉片的UWL強度下降,相對含水量下降,根系活力降低,束縛水與自由水比值上升,相對含水量與UWL之間呈高度正相關(guān),差異性顯著,束縛水與自由水比值與UWL之間呈高度負相關(guān),差異性顯著;Pn、Tr、Gs、Ci、Chla含量、Chlb含量、Chl含量下降,光合能力減弱,UWL與Pn、Tr、Gs、Ci、Chla含量、Chlb含量、Chl含量呈正相關(guān),差異性顯著;O2-產(chǎn)生速率、MDA含量、相對電導(dǎo)率上升,UWL與O2-產(chǎn)生速率、MDA含量、相對電導(dǎo)率呈負相關(guān),差異性顯著;ATP含量下降趨勢,能荷值下降,ADP和AMP含量整體上升,ATP、能荷值均與UWL呈高度正相關(guān),差異性極顯著。整個脅迫過程中,H2O2處理加劇了相對含水量、Pn、Tr、Gs、Ci、Chla含量、Chlb含量、Chl總含量以及ATP含量的降低,加劇了束縛水與自由水比值、O2-產(chǎn)生速率、MDA含量、相對電導(dǎo)率的上升,同時加劇了 UWL的下降;苯甲酸鈉處理減緩了相對含水量、Pn、Tr、Gs、Ci、Chla含量、Chlb含量、Chl總含量以及ATP含量的降低,減緩了束縛水與自由水比值、O2-產(chǎn)生速率、MDA含量、相對電導(dǎo)率的上升,同時減緩了 UWL的下降。進一步說明光合作用,能量相關(guān)與UWL產(chǎn)生有關(guān),活性氧是通過間接作用影響UWL。
[Abstract]:In this experiment, Sedum was used as the experimental material to carry out salt reverse stress, and under salt stress, reactive oxygen free radical induction and scavenging of active oxygen free radical were carried out. The changes of reactive oxygen species system, energy level and photosynthesis of ultraweak luminescent (UWL), under salt stress and free radical scavenging of reactive oxygen species were studied. To study the changes of UWL in Sedum plants under salt stress and the mechanism of production of active oxygen species (Ros). The main results of this study are as follows: 1. The changes of UWL, growth vigor, photosynthesis, reactive oxygen species system and energy level in leaves of Sedum dejingensis under NaCl salt stress were studied. The results showed that under salt stress, the UWL intensity, relative water content, root activity and the ratio of bound water to free water were decreased, and the relative water content was positively correlated with UWL, and the difference was significant. The ratio of bound water to free water was negatively correlated with UWL, and the difference was significant. Pn,Tr,Gs,Ci,Chla content, Chlb content, total Chl content decreased, photosynthetic ability decreased, UWL and Pn,Tr,Gs,Ci,Chla content, Chlb content, Chl content were highly positive correlation, the difference was significant. O _ 2-production rate, MDA content, relative conductivity increased, UWL and O _ 2- production rate, MDA content, relative conductivity were highly negative correlation, the difference was significant. ATP content decreased, energy charge value decreased, ADP and AMP content increased as a whole. ATP, energy charge value was highly positively correlated with UWL, and the difference was very significant. It reflects the degree of salt stress. The results showed that photosynthesis and energy correlation were related to UWL production, and reactive oxygen species were not the direct source of UWL production. 2. The effects of hydrogen peroxide (H2O2) and sodium benzoate on the changes of UWL, growth activity, photosynthesis, reactive oxygen system and energy level in leaves of Sedum were studied. The results showed that under salt stress, the UWL intensity, relative water content, root activity and ratio of bound water to free water of leaves treated with NaCl (control), NaCl H2O2 and NaCl benzoate) decreased, relative water content decreased, and the ratio of bound water to free water increased. The relative water content was highly positively correlated with UWL and the difference was significant. The ratio of bound water to free water was highly negatively correlated with UWL, and the difference was significant. During the whole stress process, the relative water content, Pn,Tr,Gs,Ci,Chla content, Chlb content, total Chl content and ATP content decreased, the ratio of bound water to free water, O _ 2-production rate, MDA content were increased by H2O2 treatment. The increase of relative conductivity and the decrease of UWL; Sodium benzoate treatment slowed down the decrease of relative water content, Pn,Tr,Gs,Ci,Chla content, Chlb content, total Chl content and ATP content, slowed down the ratio of bound water to free water, O _ 2-producing rate, MDA content and relative conductivity. At the same time, the decline of UWL was slowed down. It is further explained that photosynthesis and energy correlation are related to the production of UWL, and reactive oxygen species affect UWL. indirectly.
