ZD制劑對(duì)馬肢蹄軟組織損傷作用及對(duì)VEGF和bFGF表達(dá)的影響

【摘要】:ZD制劑是治療馬非開放性軟組織損傷的經(jīng)典驗(yàn)方,為探索其作用機(jī)制,本試驗(yàn)建立馬前肢急性軟組織損傷模型,應(yīng)用血清學(xué)檢測(cè),超聲方法,紅外熱成像技術(shù),組織形態(tài)學(xué)觀察,綜合評(píng)價(jià)ZD制劑對(duì)馬急性軟組織損傷的作用效果,采用免疫組織化學(xué)方法檢測(cè)ZD制劑對(duì)VEGF、bFGF在肌肉中的表達(dá),研究結(jié)果如下:(1)用10匹健康蒙古馬隨機(jī)分成2組,選取左前肢肩部臂部肌肉,用重物砸擊,制造急性軟組織損傷模型。(2)于造模前、后2h、1d、3d、5 d、7 d、15 d、21 d分別用超聲檢查評(píng)估皮下腫脹、肌肉灰度,用紅外熱成像儀檢測(cè)損傷部位局部溫度。試驗(yàn)結(jié)果表明,早期試驗(yàn)藥物組局部溫度低于陰性對(duì)照組,后期試驗(yàn)藥物組的局部溫度顯著高于陰性對(duì)照組,表明ZD制劑對(duì)急性軟組織損傷早期能夠降低局部溫度;后期能改善局部溫度;ZD制劑對(duì)肌肉灰度恢復(fù)能力顯著優(yōu)于陽性藥物組和陰性對(duì)照組,表明ZD制劑對(duì)肌肉損傷恢復(fù)有良好促進(jìn)作用;ZD制劑對(duì)皮下消腫顯著優(yōu)于陰性對(duì)照組;(3)用20匹蒙古馬隨機(jī)分為4組,選取左前肢肩部臂部肌肉,用重物砸擊,制造急性軟組織損傷模型。于造模前、后12 h、1d、3d、5 d、7 d、15 d、21 d用取樣器進(jìn)行組織取樣,制作成石蠟切片,觀察其形態(tài)學(xué)變化。結(jié)果表明,試驗(yàn)藥物組在損傷后的修復(fù)能力優(yōu)于陽性藥物組和陰性對(duì)照組。表明ZD制劑能通過促進(jìn)成纖維細(xì)胞、毛細(xì)血管新生和肌細(xì)胞的生成來進(jìn)行組織的修復(fù)再生。(4)用免疫組織化學(xué)方法對(duì)陽性細(xì)胞分析VEGF、bFGF的表達(dá),結(jié)果表明,試驗(yàn)藥物組在損傷后VEGF、bFGF的表達(dá)優(yōu)于陰性對(duì)照組和陽性藥物組,ZD制劑可能通過提高VEGF、bFGF的表達(dá)進(jìn)行損傷的修復(fù)。結(jié)論:ZD制劑在馬急性軟組織損傷模型中能夠調(diào)節(jié)受損部位的局部溫度,促進(jìn)皮下組織消腫,調(diào)控VEGF、bFGF的表達(dá),促進(jìn)成纖維細(xì)胞、毛細(xì)血管,肌細(xì)胞再生來進(jìn)行組織修復(fù)。
[Abstract]:ZD preparation is a classical prescription for the treatment of non-open soft tissue injury in horses. In order to explore its mechanism, a model of acute soft tissue injury in horse forelimb was established, and serological examination, ultrasonic method and infrared thermal imaging were used. The effect of ZD preparation on acute soft tissue injury of horse was evaluated synthetically. The expression of VEGF bFGF in muscle was detected by immunohistochemical method. The results were as follows: 1) 10 healthy Mongolian horses were randomly divided into 2 groups. The left forelimb shoulder brachial muscle was selected, and the acute soft tissue injury model was made with heavy object. The model of acute soft tissue injury was made before the model was made, and after 2 h, 1 hour, 1 day, 3 days, 5 days, 15 days and 21 days, respectively, subcutaneous swelling and muscle gray scale were evaluated by ultrasonic examination. The local temperature of the injured area was detected by infrared thermography. The results showed that the local temperature in the early test group was lower than that in the negative control group, and the local temperature in the late trial group was significantly higher than that in the negative control group, indicating that ZD preparation could lower the local temperature in the early stage of acute soft tissue injury. ZD preparation can improve the local temperature significantly better than the positive drug group and the negative control group in the recovery of muscle grayscale. The results showed that ZD had a good promoting effect on the recovery of muscle injury. ZD preparation was significantly better than that of negative control group in treating subcutaneous swelling. 20 Mongolian horses were randomly divided into 4 groups. The left forelimb and shoulder brachial muscles were randomly divided into 4 groups, which were smashed with heavy objects. A model of acute soft tissue injury was established. Before and after 12 h, 1 d, 3 d, 5 d, 7 d, 15 d and 21 d, the tissue was sampled with a sampler to make paraffin sections, and the morphological changes were observed. The results showed that the repair ability of the experimental drug group was better than that of the positive drug group and the negative control group. The results showed that ZD could promote the regeneration of tissue by promoting fibroblast, capillary neogenesis and myocyte formation.) Immunohistochemical method was used to analyze the expression of VEGF bFGF in positive cells. The expression of VEGF bFGF in the experimental group was better than that in the negative control group and the positive drug group, which may be repaired by increasing the expression of VEGF bFGF. Conclusion in the model of acute soft tissue injury of horse, the weight ZD preparation can regulate the local temperature, promote subcutaneous tissue swelling, regulate the expression of VEGF bFGF, and promote the regeneration of fibroblasts, capillaries and myocytes for tissue repair.
