池蝶蚌多糖的生物功能活性分析

【摘要】:池蝶蚌(Hyriopsis schlegelii)屬于軟體動物們,瓣鰓綱,蚌科,帆蚌屬,原產(chǎn)于日本,是優(yōu)質(zhì)淡水育珠蚌,所產(chǎn)珍珠個體大、圓、光,品質(zhì)十分優(yōu)良。1997年從日本引進我國,由于其育珠性能良好、抗病能力強,現(xiàn)被作為優(yōu)良的經(jīng)濟水產(chǎn)品種推廣應(yīng)用。本實驗室對池蝶蚌持續(xù)研究20年,在遺傳育種、優(yōu)化培養(yǎng)等方面取得了一定成果,但對其生物學(xué)活性物質(zhì)等方面的研究還較少。池蝶蚌多糖類物質(zhì)的提取及相關(guān)功能活性的研究剛剛起步,本研究旨在對池蝶蚌多糖的生物活性做初步的研究,為池蝶蚌多糖的開發(fā)利用提供一定的依據(jù)。針對以前的多糖提取方法,本研究調(diào)整了池蝶蚌多糖的提取技術(shù),使提取更加方便,得到了池蝶蚌多糖組分HSP-1和HSP-2,提取率分別為1.9%和0.95%;對池蝶蚌多糖的抗腫瘤、免疫及抗氧化相關(guān)的活性做了初步的分析探討。在抗氧化實驗中,分別選取DPPH、羥自由基和超氧自由基的清除活性以及還原力作為體外指標(biāo);體內(nèi)抗氧化測定,給D-半乳糖小鼠衰老模型定期灌胃池蝶蚌多糖,發(fā)現(xiàn)在小鼠血清中,池蝶蚌多糖(HSP-1)高劑量組的T-SOD酶活力為155U/ml,而陰性對照組血清中則是122 U/ml,MDA陰性對照組含量為13.64nmol/ml,高劑量組的含量為9.37 nmol/ml,總抗氧化活性高劑量組為11.22U/ml,陰性對照組僅為6.48;在肝臟中,MDA陰性對照組含量為21.15 nmol/ml,高劑量組僅為11.30 nmol/ml,陰性對照組的T-SOD活力為71.55U/mg,高劑量組為134.33 U/mg,總抗氧化能力陰性對照組為4.43 U/mg,高劑量組為13.64 U/mg。經(jīng)過實驗結(jié)果數(shù)據(jù)分析比較可以看出HSP-1具有提高抗氧化酶活力及降低氧化產(chǎn)物MDA含量的能力,抗氧化實驗結(jié)果表明池蝶蚌多糖HSP-1具有較好的抗氧化功能。選取SMMC-7721和SGC-7901兩株腫瘤細胞株,用HSP-1和HSP-2進行了抗腫瘤生物活性實驗,100 mg/L時HSP-1、HSP-2及可溶性淀粉對SMMC-7721細胞增殖影響效果分別為1.11±0.07、0.98±0.13和1.06±0.06,對于SGC-7901細胞影響為0.94±0.03、1.09±0.08和0.85±0.07,結(jié)果顯示,兩種多糖組分在體外均未表現(xiàn)出明顯抑制腫瘤的結(jié)果;而另一方面,將池蝶蚌多糖體外作用于小鼠脾臟淋巴細胞時,結(jié)果顯示其能夠使脾臟淋巴細胞增殖,脾臟顯著增大,高劑量組脾重指數(shù)約為0.358,而陰性對照組的脾重指數(shù)約為0.099,因此我們推測池蝶蚌多糖具有提高機體免疫功能的活性。
[Abstract]:Hyriopsis Schlegelii) is a kind of mollusk. It is native to Japan. It is a kind of high-quality freshwater oyster. The pearls produced by Hyriopsis schlegelii are of great size, round, light and excellent quality. In 1997, China was introduced from Japan because of its good bead breeding performance.Strong resistance to disease, is being used as an excellent economic aquatic varieties to promote and use.In our laboratory, we have made some achievements in genetics and breeding, optimization of culture and so on, but there are few studies on biological active substances in this laboratory.The extraction of polysaccharides and their related functional activities have just started. The purpose of this study is to make a preliminary study on the biological activities of polysaccharides from the mussels, and to provide some basis for the development and utilization of polysaccharides.In view of the previous polysaccharide extraction methods, this study adjusted the extraction technology of Polysaccharide from Phellodendron ponyrii to make the extraction more convenient, and obtained polysaccharide components HSP-1 and HSP-2, the extraction rates were 1.9% and 0.95%, respectively.The activities related to immunity and anti-oxidation were preliminarily analyzed and discussed.The scavenging activity and reductive capacity of DPPH, hydroxyl radical and superoxide free radical were selected as the in vitro indexes respectively.It was found that in mouse serum,The activity of T-SOD enzyme was 155U / ml in high dose group, 13.64nmol / ml in negative control group, 9.37nmol / ml in high dose group, 11.22Umlml in high dose group and 6.48nmol / ml in negative control group.The content of T-SOD in liver was 21.15 nmol / ml in negative control group, 11.30 nmol / ml in high dose group, 71.55 U / mg in negative control group, 134.33 U / mg in high dose group, 4.43 U / mg in total antioxidant capacity negative control group and 13.64 U / mg in high dose group.The results showed that HSP-1 could increase the activity of antioxidant enzymes and decrease the content of MDA. The results of antioxidant experiments showed that HSP-1 had better antioxidant function.On the other hand, when the polysaccharides were treated in vitro with the mouse spleen lymphocytes, the results showed that the two polysaccharides could make the spleen lymphocytes proliferate and the spleen increase significantly.The spleen weight index of the high dose group was about 0.358, while that of the negative control group was about 0.099. Therefore, we speculated that the polysaccharide from the mussel had the activity of improving the immune function of the body.
