三種野生食藥用真菌的馴化和培養(yǎng)特性研究

【摘要】:茶呰子葉狀層菌、粉托鬼筆、短裙竹蓀均是野生食藥用真菌。據(jù)研究,茶呰子葉狀層菌與金銀花的功效有相似之處,可用于消炎、解毒、止痛等,目前已被制成保健品制劑,對各類炎癥及感冒有極好的療效。粉托鬼筆首先由本實(shí)驗(yàn)室進(jìn)行研究,在本實(shí)驗(yàn)室中已栽培出子實(shí)體,但尚未進(jìn)行大規(guī)模的栽培。短裙竹蓀是一種味道極佳的食藥用真菌,具有降血脂,調(diào)節(jié)脂肪酸預(yù)防高血壓的作用。目前,這三種野生食藥用真菌采摘量有限,尚未進(jìn)行過系統(tǒng)的菌絲培養(yǎng)研究,也限制了它們的藥理研究。本論文進(jìn)行了茶呰子葉狀層菌、粉托鬼筆的菌種分離及鑒定、固體培養(yǎng)特性和液體發(fā)酵培養(yǎng)條件及菌絲化學(xué)成分含量研究。短裙竹蓀的固體培養(yǎng)和發(fā)酵罐發(fā)酵培養(yǎng)條件及菌絲化學(xué)成分含量研究。為以后進(jìn)行大規(guī)模的工業(yè)發(fā)酵生產(chǎn)做好準(zhǔn)備工作。首先對采集的野生茶呰子葉狀層菌子實(shí)體進(jìn)行菌種分離及菌種鑒定,確定組織分離所得菌絲體即為茶呰子葉狀層菌菌絲體。采用固體皿培法研究固體培養(yǎng)特性:發(fā)現(xiàn)茶呰子葉狀層菌菌絲體在麥麩、麥芽汁、胡蘿卜、PDA、GPC、GPY固體培養(yǎng)基上均能生長,在麥芽汁固體培養(yǎng)基上生長速度最快,在第50天時(shí)的麥麩固體培養(yǎng)基上生物量最大,在第40天時(shí)GPY培養(yǎng)基上的單位質(zhì)量的菌絲體中三萜含量為0.39%,考慮獲得三萜的總量最高,應(yīng)選擇60天的麥麩培養(yǎng)基;茶呰子葉狀層菌對碳源和有機(jī)氮源的適應(yīng)性較強(qiáng),在麥芽糖、果糖、木糖、乳糖上生長較好,除了在尿素中不能生長,在其他氮源中均能生長。采用搖瓶發(fā)酵法篩選發(fā)酵培養(yǎng)基:茶呰子葉狀層菌在只有碳源+氮源+生長因子的培養(yǎng)液中也能生長,但在麥芽汁培養(yǎng)液中生長最好,麥芽汁培養(yǎng)液適合作為茶呰子葉狀層菌的種子培養(yǎng)液和發(fā)酵培養(yǎng)液;在搖瓶發(fā)酵中,三萜含量不低于1%,多糖含量在15%以上,第7天的生物量達(dá)到最大,同時(shí)采用HPLC測得腺苷獲得量最多。隨著液體發(fā)酵時(shí)間的延長,茶呰子葉狀層菌菌球顏色逐漸由乳白色變?yōu)樽睾稚?且發(fā)酵液顏色也逐漸變?yōu)樽睾稚?并有一股清新的果香味。本研究采用組織分離法對粉托鬼筆菌種進(jìn)行菌種分離并進(jìn)行分子生物學(xué)鑒定得出分離所得菌絲體為粉托鬼筆。粉托鬼筆在不同固體培養(yǎng)基上均能生長,在麥麩、麥芽汁、胡蘿卜、PDA固體培養(yǎng)基上生長較好,粉托鬼筆對碳源和氮源的適應(yīng)性較好,在不同碳源、氮源條件下,其菌絲的生長速度和氣生菌絲的形態(tài)有較大差異。通過不同培養(yǎng)液及不同碳源及氮源對液體發(fā)酵培養(yǎng)基的篩選發(fā)現(xiàn):PDA培養(yǎng)液適合作為粉托鬼筆的種子培養(yǎng)液,在10L的發(fā)酵罐中投料7L,蔗糖140g,大豆蛋白胨42g,酵母浸粉70g,接種量5%,溫度26℃,轉(zhuǎn)速150rpm,通氣量1:0.3,培養(yǎng)至58h適合放罐。菌絲體中多糖含量達(dá)18.02%。適合作為判斷放罐標(biāo)準(zhǔn)的指標(biāo)是菌絲生物量及發(fā)酵液總糖含量。短裙竹蓀菌絲體在所試不同培養(yǎng)上均能生長,在GPY、PDA、麥芽汁、麥麩、胡蘿卜固體培養(yǎng)基上生長較好,在GPY固體培養(yǎng)基上生長速度最快,生物量最大,為145.50mg。短裙竹蓀對碳源和有機(jī)氮源的適應(yīng)性很強(qiáng)。短裙竹蓀液體種子培養(yǎng)基為PDA培養(yǎng)液,在10L的發(fā)酵罐中投料7L,蔗糖140g,大豆蛋白胨42g,酵母浸粉70g,接種量5%,溫度26℃,轉(zhuǎn)速150rpm,通氣量1:0.2,培養(yǎng)至90h適合放罐。菌絲體中多糖含量達(dá)17.92%。
