白檀組織培養(yǎng)技術研究

【摘要】:白檀(Symplocos paniculata)為山礬科(Symplocaceae)山礬屬落葉灌木或小喬木,花朵潔白繁多,亮藍果實密布樹冠,樹形優(yōu)美,具有較高的園林觀賞價值。同時,全株在工、醫(yī)、農(nóng)業(yè)等方面均有廣泛用途,是集觀賞價值和經(jīng)濟價值于一身的優(yōu)良野生資源。為了能夠更好地開發(fā)、保護和綜合利用白檀野生資源,高效、穩(wěn)定的白檀組織培養(yǎng)技術體系亟待建立。本文以白檀不同部位的帶有腋芽的莖段和種子為外植體,對外植體消毒、啟動培養(yǎng)、增殖培養(yǎng)、生根培養(yǎng)、煉苗移栽的影響因素進行研究,建立了白檀組織培養(yǎng)體系。主要研究結(jié)果如下:1.帶腋芽嫩莖為最適宜的外植體,最佳滅菌方案為0.1%升汞溶液滅菌3分鐘,污染率可控制在18.2%;白檀胚做外植體,最佳預處理為浸泡在1100mg/L GA3溶液中24h,經(jīng)75%酒精浸泡30s,再用0.1%升汞溶液滅菌7min,可將污染率控制在9.2%。2.嫩莖的最佳啟動培養(yǎng)基為1/2MS+0.5mg/L 6-BA+0.1mg/L NAA,啟動率達87.44%;白檀胚經(jīng)預處理及滅菌處理后,接種到1/2WPM培養(yǎng)基,啟動率達78.59%。3.采用叢生芽增殖途徑對白檀試管苗進行繼代增殖培養(yǎng),試管苗最適增殖培養(yǎng)基為WPM+1.7mg/L ZT,平均增殖系數(shù)可達5.03,芽平均高度達5.00cm。4.雙芽莖段最適做白檀試管苗增殖材料;25℃/15℃(晝12h/夜12h)溫差處理可使白檀試管苗生長健壯。5.白檀試管苗生根最適培養(yǎng)基為WPM+0.5mg/L IBA+0.5mg/L NAA+20g/L蔗糖+2g/L活性炭+8g/L瓊脂,生根率達93%,平均生根數(shù)量為6根。6.白檀試管苗最適移栽基質(zhì)為體積比1:1的園土和草炭土混合基質(zhì),移栽成活率為83%。
[Abstract]:Symplocos paniculata) is a deciduous shrub or small tree of the genus Symplocaceae. It has a wide range of white flowers, bright blue fruit, dense crown, beautiful tree shape and high ornamental value in gardens. At the same time, the whole plant is widely used in engineering, medicine, agriculture and so on. It is an excellent wild resource with both ornamental value and economic value. In order to develop, protect and make comprehensive use of the wild resources of white sandalwood, it is highly effective. It is urgent to establish a stable tissue culture system. In this paper, the stem segments and seeds with axillary buds were used as explants to sterilize the explants, start culture, proliferate culture, and root culture. The main results are as follows: 1. The young stem with axillary bud is the most suitable explant, and the best sterilization scheme is 0.1% mercuric solution for 3 minutes. The pollution rate can be controlled at 18.2cm, and the embryo is used as explant. The best pretreatment was immersion in 1100mg / L GA3 solution for 24h, soaking in 75% alcohol for 30s, sterilizing with 0.1% mercuric solution for 7 mins. The pollution rate was controlled at 9.2 mg / L. The best starting medium for tender stem was 1 / 2ms 0.5 mg / L 6-BA 0.1 mg / L NAA, the priming rate was 87.444.After pretreatment and sterilization, the best starting medium for the tender stem was 87.444.After pretreatment and sterilization, the best starting medium for the tender stem was: 1 / 2 MS, 0.5 mg / L 6-BA 0.1 mg / L NAA. When inoculated to 1 / 2 WPM medium, the initiation rate was 78.59.3.The propagation pathway of cluster buds was used to subculture the test-tube plantlets of sandalwood. The optimum culture medium was WPM 1.7 mg / L ZT, the average multiplication coefficient was 5.03, and the average height of buds was 5.00 cm. 4.
