中國(guó)野生大豆中大豆花葉病毒的鑒定及演化分析

【摘要】:大豆花葉病毒(Soybean mosaic virus,SMV)是一種世界性的大豆病害,嚴(yán)重影響大豆的產(chǎn)量和品質(zhì)。目前,對(duì)該病毒的調(diào)查主要在栽培大豆中展開,而對(duì)中國(guó)野生大豆所攜帶的SMV種類和遺傳多樣性的研究尚不充分。本研究對(duì)2013年采集自中國(guó)東北和南方地區(qū)共60份野生大豆種子攜帶病毒的情況進(jìn)行了調(diào)查。結(jié)果表明,其中12份野生大豆材料具有典型花葉、皺縮等癥狀。通過(guò)設(shè)計(jì)兩對(duì)馬鈴薯Y病毒屬(Potyvirus)兼并引物(PVYG-F1/R1和PVYG-F2/R2)進(jìn)行RT-PCR檢測(cè)及測(cè)序分析,這12個(gè)分離物均為SMV,并可以歸類為9種不同的序列類型;對(duì)其中7種SMV類型,成功進(jìn)行了全基因組測(cè)序。將這7條SMV全基因組新序列與NCBI數(shù)據(jù)庫(kù)中的83條SMV基因組序列共同排序并構(gòu)建系統(tǒng)演化樹后,結(jié)果發(fā)現(xiàn),分離于南方野生大豆材料中的4條SMV全基因組序列位于演化支H中,與已經(jīng)報(bào)道的SMV分離物SC7、BYX006和LJZ002親緣關(guān)系相近。這4個(gè)新發(fā)現(xiàn)的SMV分離物均屬于重組型SMV,其5'端含有一段大約900 bp的菜豆普通花葉病毒(Bean common mosaic virus,BCMV)的基因序列。而分離于北方野生大豆材料中的3條SMV全基因組序列分別位于演化支Ⅲ和Ⅳ中。由此可知,中國(guó)野生大豆也是SMV的重要宿主,在SMV的傳播過(guò)程中起著重要作用。本研究進(jìn)而選取了南、北方SMV分離物SC7-N與HLJBADS001,對(duì)所采集野生大豆材料中代表性個(gè)體的繁殖后代進(jìn)行了初步侵染實(shí)驗(yàn)。結(jié)果顯示,33份樣品抗HLJBADS001與SC7-N兩個(gè)SMV分離物,26份樣品只抗HLJBADS001。因此,野生大豆中蘊(yùn)藏著豐富的抗SMV的資源。
[Abstract]:Soybean mosaic virus (Soy bean mosaic virus) is a worldwide soybean disease, which seriously affects the yield and quality of soybean. However, the SMV species and genetic diversity of wild soybean in China were not fully studied. In 2013, 60 wild soybean seed carriers from Northeast and South China were investigated. Twelve wild soybean materials had typical mosaic, shrinkage and other symptoms. By designing two pairs of potato Y virus degenerate primers, PVYG-F1 / R1 and PVYG-F2 / R2), RT-PCR detection and sequencing were performed. The 12 isolates are SMVs and can be classified into 9 different sequence types. The whole genome was sequenced successfully. The seven new SMV genome sequences were sequenced with 83 SMV genome sequences in the NCBI database and the phylogenetic tree was constructed. Four SMV complete genome sequences isolated from southern wild soybean were located in the evolutionary branch H. The four newly discovered SMV isolates all belong to recombinant SMVs, and their 5'ends contain a gene sequence of about 900bp common bean mosaic virus, common mosaic virusBCMV, which is similar to the reported SMV isolates SC7BYX006 and LJZ002.
