利用cDNA-AFLP技術(shù)分析全膜覆蓋對玉米苗期基因表達(dá)的影響

【摘要】:為了研究玉米苗期在全膜覆蓋條件下,基因的差異表達(dá)。本實(shí)驗(yàn)利用cDNA-AFLP技術(shù)對全膜覆蓋條件下種植的玉米與不覆膜條件下種植的玉米,進(jìn)行差異表達(dá)基因片段的篩選、分離、測序和功能性分析,結(jié)果如下:1.建立適合玉米苗期根系和葉片總RNA提取的方法,并將總RNA經(jīng)過反轉(zhuǎn)錄形成雙鏈cDNA。使用PstI和MseI兩種內(nèi)切酶對雙鏈cDNA進(jìn)行酶切,設(shè)計(jì)與兩種內(nèi)切酶相對應(yīng)的接頭以及9對選擇性擴(kuò)增引物,進(jìn)行特異片段擴(kuò)增及測序。2.運(yùn)用cDNA-AFLP技術(shù),共擴(kuò)增出條帶1481條,336條差異表達(dá)片段,約占總條帶數(shù)的22.7%。隨后將其中的60條進(jìn)行測序得到58條測序結(jié)果。3.利用NCBI系統(tǒng)對58條測序結(jié)果進(jìn)行同源性分析和功能預(yù)測。結(jié)果如下:與植物同源性較高的有13條并且大多具有蛋白功能;與動物同源性較高的有6條;與微生物同源性較高的有20條;另有,19條差異表達(dá)片段未能找到同源性序列。4.獲得的與植物同源性較高的13條基因片段中,有3條與小麥、高粱、柑桔植物體中相關(guān)功能基因高度相似,其余10條均與玉米體內(nèi)某些克隆基因高度相似。以上這些差異條帶的獲得為更深層次研究玉米基因在覆膜條件下的差異表達(dá)打下了良好的基礎(chǔ)。5.測序結(jié)果中獲得了 20條與微生物同源性高度相似的基因片段,這些基因片段的獲得以及在玉米生長過程中的作用還需進(jìn)一步研究確定。
[Abstract]:In order to study the differential expression of genes in seedling stage of maize under the condition of full film mulching, cDNA-AFLP technique was used to treat maize planted under the condition of full film mulching and maize without film mulching. Screening, isolation, sequencing and functional analysis of differentially expressed gene fragments were carried out. The results were as follows: 1. A suitable method for extracting total RNA from roots and leaves of maize at seedling stage was established. The total RNA was reversed to form double-stranded cDNA.The double strand cDNA was digested by PstI and MseI endonuclease. The specific fragments were amplified and sequenced by 9 pairs of primers. 1 481 bands were amplified by cDNA-AFLP technique. 336 differentially expressed fragments. Then 60 of them were sequenced to get 58 sequencing results .3.The homology analysis and functional prediction of 58 sequencing results were carried out by using NCBI system. The results were as follows:. Below:. 13 of them had high homology with plants and most of them had protein function. Six of them had high homology with animals. There were 20 high homology with microorganisms. Another 19 differentially expressed fragments could not find the homologous sequence. 4. Of the 13 genes with high homology with plants, 3 were with wheat and sorghum. The related functional genes in citrus plants are highly similar. The other 10 genes were highly similar to some cloned genes in maize. The above results laid a good foundation for further research on the differential expression of maize genes under the condition of film mulching. 5. Sequencing results. In the. There were 20 gene fragments with high homology with microorganism. The acquisition of these gene fragments and their roles in maize growth need to be further studied.
