復(fù)合益生菌對(duì)羅非魚免疫相關(guān)基因的影響

【摘要】:本研究通過把三種復(fù)合益生菌添加到飼料中,分別飼喂羅非魚(Tilapia),確定提升免疫保護(hù)率最佳的復(fù)合益生菌種類,并從分子水平初步探索復(fù)合益生菌對(duì)羅非魚免疫力影響的作用機(jī)理。試驗(yàn)共分4組,3個(gè)試驗(yàn)組和1個(gè)對(duì)照組。3個(gè)試驗(yàn)組用分別添加了等比例的地衣芽孢桿菌(Bacillus licheniformis)+枯草芽孢桿菌(Bacillus subtilis)、地衣芽孢桿菌(Bacillus licheniformis)+凝結(jié)芽孢桿菌(Bacillus coagulans)、地衣芽孢桿菌(Bacillus licheniformis)+糞腸球菌(Enterococcus faecalis)的飼料飼喂羅非魚,添加量為2‰。飼喂40天,期間每隔10天取羅非魚肝臟、腎臟、頭腎、脾臟以及鰓五個(gè)器官,采用實(shí)時(shí)熒光定量PCR技術(shù)檢測各器官中環(huán)氧化酶-2(COX-2)、白細(xì)胞介素-1β(IL-1β)和腫瘤壞死因子-a (TNF-a)這三種免疫相關(guān)基因的相對(duì)表達(dá)量變化情況。40天后,以攻毒試驗(yàn)來檢測復(fù)合益生菌對(duì)羅非魚抗感染保護(hù)率的影響。分別給羅非魚胸鰭基部注射嗜水氣單胞菌、維氏氣單胞菌和無乳鏈球菌三種羅非魚常見致病菌。試驗(yàn)結(jié)果表明:三種不同的復(fù)合益生菌都顯著(P0.05)誘導(dǎo)羅非魚五個(gè)器官中CoX-2、IL-1β和TNF-a基因的表達(dá),其中地衣枯草組誘導(dǎo)表達(dá)效果最明顯;基因誘導(dǎo)表達(dá)在免疫器官腎臟、頭腎和脾臟中要比非免疫器官肝和鰓中明顯;地衣枯草組提高了羅非魚對(duì)嗜水氣單胞菌(Aeromonas hydrophila)、維氏氣單胞菌(Aeromonas veronii)和無乳鏈球菌(Streptococcus agalactiae)的抗感染保護(hù)率;地衣凝結(jié)組提高了羅非魚對(duì)嗜水氣單胞菌和維氏氣單胞菌的抗感染保護(hù)率。
