【摘要】:枸杞作為寧夏經(jīng)濟(jì)作物的支柱,倍受政府的關(guān)注。但是,各種自然病菌、病蟲(chóng)害以及環(huán)境和人為等因素,直接或者間接地影響了寧夏枸杞的果實(shí)質(zhì)量和產(chǎn)量,其中枸杞根腐病病菌對(duì)枸杞的影響最為嚴(yán)重。牛心樸子是生長(zhǎng)于寧夏荒漠半荒漠地區(qū)的有毒植物,項(xiàng)目組前期的研究成果表明其生物堿類生成具有抗真菌的作用。本論文為了從牛心樸子中尋找能抑制尖孢鐮刀菌的活性成分,對(duì)牛心樸子的生物堿進(jìn)行了系統(tǒng)分離,在獲得初步抑制活性的前提下,采用pH區(qū)帶精制逆流色譜技術(shù)大量制備有效成分,對(duì)分離條件進(jìn)行探討。主要內(nèi)容如下:根據(jù)項(xiàng)目組前期研究成果,論文以牛心樸子中的菲并吲哚里西丁類生物堿為分離目標(biāo),采用經(jīng)典色譜方法和核磁共振技術(shù),從牛心樸子總生物堿中分離了3個(gè)化合物,鑒定了兩個(gè)化合物的結(jié)構(gòu),分別為7-demethoxylophorine(antofine)和一個(gè)脂鏈化合物,初步測(cè)定了鮮牛心樸子中生物堿的含量為1.63%。采用菌絲生長(zhǎng)抑制法對(duì)總生物堿和antofine抑制尖孢鐮刀菌的生長(zhǎng)活性進(jìn)行了初步研究結(jié)果表明,高濃度(40μg/mL和50μg/mL)下,總生物堿表現(xiàn)出與市售抗根腐病化學(xué)農(nóng)藥相近的抑制活性,提示有進(jìn)一步開(kāi)發(fā)為抗枸杞根腐病的植物源農(nóng)藥的潛力。值得注意的是,antofine在較低濃度(20μg/m L)下,就表現(xiàn)出良好的抑制尖孢鐮刀菌生長(zhǎng)的活性,且優(yōu)于市售化學(xué)農(nóng)藥百菌清和多菌靈,antofine可以作為新的抗根腐病的植物源農(nóng)藥或先導(dǎo)化合物進(jìn)行進(jìn)一步的研究。pH區(qū)帶精制逆流色譜是一種液液分配色譜。為了富集和純化antofine,以正己烷-乙酸乙酯-甲醇-水為溶劑體系,以溶劑在儀器中的保留率和antofine的得率作為指標(biāo),篩選了保持劑的加入量、儀器轉(zhuǎn)速、流動(dòng)相流速等分離條件,優(yōu)化出最佳分離條件為溶劑體系正己烷:乙酸乙酯:甲醇:水(3:6:1:5.5),儀器轉(zhuǎn)速650r/min、流速2mL/min。以此條件,一次制備得到antofine 44mg,純度90%。
[Abstract]:Lycium barbarum, as the pillar of economic crops in Ningxia, has attracted the attention of the government. However, various natural pathogens, pests and diseases, environmental and human factors, directly or indirectly affect the fruit quality and yield of Lycium barbarum in Ningxia. The pathogen of Lycium barbarum root rot has the most serious effect on Lycium barbarum. Prunus chinensis is a poisonous plant growing in the desert and semi-desert area of Ningxia. The previous research results of the project group showed that its alkaloids had antifungal effect. In order to search for the active components which could inhibit Fusarium oxysporum in this paper. The alkaloids of Prunus chinensis were systematically separated. On the premise of obtaining the primary inhibitory activity, a large number of effective components were prepared by using pH zone refining countercurrent chromatography technique. The main contents are as follows: according to the previous research results of the project group, the separation of phenanthrene indorisidine alkaloids from Prunus Niuxinensis was taken as the separation target. Three compounds were isolated from total alkaloids of Prunus chinensis by classical chromatography and nuclear magnetic resonance (NMR) technique, and the structures of two compounds were identified. They were 7-demethoxylophorine antofine) and a lipid chain compound. The content of alkaloid was 1.63. The activity of total alkaloids and antofine against Fusarium oxysporum was studied by hyphal growth inhibition method. . At high concentrations of 40 渭 g / mL and 50 渭 g / mL, the total alkaloids showed similar inhibitory activities as those of the marketed chemical pesticides against root rot. It is suggested that there is potential for further development of plant-based pesticides against Lycium barbarum root rot. It is worth noting that antofine is at a lower concentration of 20 渭 g / m ~ (L). It showed good activity in inhibiting the growth of Fusarium oxysporum and was superior to chlorothalonil and carbendazim in the market. Antofine can be further studied as a new phytochemical or lead compound against root rot. Ph zone refining countercurrent chromatography is a liquid-liquid partition chromatography. In order to enrich and purify anto. Fine. Using n-hexane ethyl acetate methanol-water as solvent system the retention rate of solvent in the instrument and the yield rate of antofine were used as indexes to select the amount of retention agent and the rotational speed of the instrument. The optimum separation conditions were as follows: solvent system: n-hexane: ethyl acetate: methanol: water: 3: 6: 1: 5.5. 5. The rotational speed of the instrument was 650 r / min. The flow rate was 2 mL / min. Under this condition, 44 mg antofine was prepared at one time, and the purity was 90 mg.
