miR-4312促進乳腺上皮細(xì)胞增殖并抑制其凋亡

乳腺癌是威脅女性健康的重要因素,其發(fā)病機制很復(fù)雜,受多重因素影響。近年來miRNA在腫瘤發(fā)生發(fā)展中的作用越來越受到關(guān)注。miRNA是一種內(nèi)源性非編碼小RNA,參與基因表達調(diào)控過程,腫瘤組織和細(xì)胞中常有異常表達。目前研究發(fā)現(xiàn)多種miRNA參與乳腺癌發(fā)生發(fā)展,如miR-21、miR-155、miR-182、miR-10b、miR-27a、miR-9等有致癌作用,而let-7家族、miR-200家族、miR-205、miR-335、miR-145、miR-19等有抑癌作用。但多數(shù)miRNA在乳腺癌中的作用機制尚不清楚。本文通過對比正常人和乳腺癌患者血清中miRNA豐富度,發(fā)現(xiàn)一系列miRNA在腫瘤患者血清中的差異表達,并對其用qPCR技術(shù)進行了驗證。除了文獻已報導(dǎo)的miRNA外,本文發(fā)現(xiàn)miR-4312在乳腺癌患者血清和乳腺癌細(xì)胞中都高表達。在MCF-10A細(xì)胞中過表達miR-4312,發(fā)現(xiàn)細(xì)胞生長曲線斜率明顯升高;G0/G1期細(xì)胞明顯減少,而G2/M期細(xì)胞明顯增多,凋亡細(xì)胞數(shù)量明顯減少,細(xì)胞遷移能力增強。同時還發(fā)現(xiàn)解旋酶MCM2表達水平提升,細(xì)胞內(nèi)凋亡相關(guān)蛋白Caspase 3、BCL2、...
