IGF2BP2(IMP2)是小鼠早期合子基因組激活的關(guān)鍵調(diào)控因子

背景:哺乳動(dòng)物配子受精前,其基因組處于無(wú)轉(zhuǎn)錄活性狀態(tài);經(jīng)過(guò)減數(shù)分裂形成的卵子和精子在受精過(guò)程后相互識(shí)別并結(jié)合,遺傳物質(zhì)融合并形成合子,進(jìn)一步發(fā)育成新的個(gè)體。受精卵基因表達(dá)受到合子基因組激活(ZGA)的調(diào)控,此前轉(zhuǎn)錄沉默的基因組進(jìn)行重編程,建立其特有的全能性,并進(jìn)一步激活早期胚胎發(fā)育所需的基因表達(dá)。ZGA是細(xì)胞核重編程的過(guò)程,可使合子基因組從受精時(shí)的轉(zhuǎn)錄沉默狀態(tài)轉(zhuǎn)變?yōu)楹笃诘募せ顮顟B(tài),合子基因的表達(dá)產(chǎn)物能夠逐漸替代卵母細(xì)胞中貯存的母源因子維持早期胚胎發(fā)育,是哺乳動(dòng)物及其他脊椎動(dòng)物早期胚胎發(fā)育的關(guān)鍵階段。不同物種ZGA的發(fā)生時(shí)間不同,在小鼠中ZGA開始于2細(xì)胞期,而人類則為4-8細(xì)胞期。這個(gè)合子發(fā)育過(guò)程中基因表達(dá)模式的劇變均發(fā)生在轉(zhuǎn)錄和翻譯水平,基因組激活的差異可能導(dǎo)致的不同結(jié)果,并對(duì)哺乳動(dòng)物的妊娠結(jié)局產(chǎn)生一定影響。根據(jù)功能和轉(zhuǎn)錄活性可將ZGA分為次要和主要兩個(gè)階段:起始階段即次要ZGA,發(fā)生在1細(xì)胞胚胎期的S相到2細(xì)胞期的G1相,為緩慢發(fā)育階段;而主要ZGA發(fā)生在2細(xì)胞晚期,大量轉(zhuǎn)錄過(guò)程快速發(fā)生,基因調(diào)控機(jī)制開始運(yùn)行。兩個(gè)階段的正確激活和調(diào)節(jié)對(duì)于基因的表達(dá)至關(guān)重要,并在細(xì)胞增殖和早期胚胎發(fā)...
