【摘要】：乳腺癌易感基因1(Breast cancer 1,early onset, BRCAI)突變是導(dǎo)致人家族型乳腺癌發(fā)生的主要原因之一。但是關(guān)于犬BRCA1在乳腺腫瘤發(fā)生中的作用,研究尚不深入。在這篇論文中,檢測了犬乳腺腫瘤中BRCA1啟動(dòng)子區(qū)的突變情況,mRNA和蛋白的表達(dá)情況,編碼區(qū)的突變情況和BRCA1啟動(dòng)子區(qū)的甲基化情況。并希望通過這些實(shí)驗(yàn),進(jìn)一步闡明BRCA1在犬乳腺腫瘤發(fā)生中的作用。由于犬BRCA1啟動(dòng)子序列尚沒有確定,首先通過生物信息學(xué)的方法預(yù)測了犬BRCA1基因的啟動(dòng)子序列,隨后用啟動(dòng)子活性分析的方法進(jìn)行驗(yàn)證。研究結(jié)果顯示,預(yù)測的犬BRCA1啟動(dòng)子組的報(bào)告基因活性明顯高于兩個(gè)陰性對照組(P0.01)。為了檢測犬乳腺腫瘤中BRCA1啟動(dòng)子區(qū)的突變情況,從中國農(nóng)業(yè)大學(xué)動(dòng)物醫(yī)院獲取犬乳腺腫瘤組織樣本,根據(jù)病理特征將所得的樣本分成良性乳腺腫瘤組和惡性乳腺腫瘤組,每組均有15例組織樣本,并川10例犬止常乳腺組織作為對照組。通過PCR產(chǎn)物測序,分析了各組組織中BRCA1啟動(dòng)子區(qū)的突變情況。啟動(dòng)子測序結(jié)果顯示,在正常乳腺組織和良性乳腺腫瘤中,均沒有檢測到啟動(dòng)子區(qū)的突變；在惡性乳腺腫瘤中,在啟動(dòng)子區(qū)發(fā)現(xiàn)一個(gè)19959098Cdel的點(diǎn)突變,突變率為46.7%(7/15)。為了檢測犬乳腺腫瘤中BRCA1表達(dá)的改變,分別通過定量PCR和免疫組織化學(xué)的方法,檢測上述各組組織中BRCA1基因mRNA和蛋白的表達(dá)情況。研究結(jié)果顯示,在犬的良性和惡性乳腺腫瘤中,BRCA1的mRNA表達(dá)量明顯低于正常組織的對照組(P0.05)；BRCA1蛋白的表達(dá)量,惡性乳腺腫瘤組明顯低于正常乳腺組織的對照組(P0.05)。為了檢測犬乳腺腫瘤中BRCA1編碼區(qū)的突變情況和啟動(dòng)子區(qū)的甲基化情況,分別用PCR產(chǎn)物測序和亞硫酸氫鹽測序的方法,檢測了上述各組組織中,BRCA1編碼區(qū)的突變情況和啟動(dòng)子區(qū)的甲基化情況。研究結(jié)果顯示,在正常乳腺組織中,沒有檢測到BRCA1啟動(dòng)子區(qū)發(fā)生突變；在乳腺腫瘤組織中,檢測到BRCA1編碼區(qū)的4個(gè)突變位點(diǎn),其中3個(gè)為新發(fā)現(xiàn)的突變位點(diǎn)。在正常乳腺組織和良性乳腺腫瘤組織DNA樣本中,均沒有檢測到BRCA1啟動(dòng)子區(qū)的甲基化；在1例惡性乳腺乳腺腫瘤組織DNA樣本中,檢測到BRCA1啟動(dòng)子區(qū)發(fā)生甲基化。綜上所述,在犬惡性乳腺腫瘤中,檢測到位于BRCA1啟動(dòng)子區(qū)的一個(gè)高突變率點(diǎn)突變位點(diǎn)。在犬良性和惡性乳腺腫瘤中,BRCA1 mRNA的水平有明顯的下調(diào)。在犬惡性乳腺腫瘤中,BRCA1蛋白的水平有明顯的下調(diào)。在犬的乳腺腫瘤中,檢測到位于編碼區(qū)的3個(gè)新的點(diǎn)突變位點(diǎn)。在犬的惡性乳腺腫瘤中,檢測到BRCA1啟動(dòng)子區(qū)發(fā)生甲基化。
[Abstract]:Breast cancer susceptibility gene 1 (Breast cancer 1, early onset, BRCAI) mutation is a major cause of human familial breast cancer. But on the role of canine BRCA1 in breast tumors, has not been studied in detail. In this paper, the detection of mutations in BRCA1 promoter region of canine breast tumor the expression of mRNA and protein, encoding region mutation and hypermethylation of the BRCA1 promoter region. And hope that through these experiments, to further elucidate the role of BRCA1 in canine mammary tumors. The canine BRCA1 promoter sequence has not been established, first by bioinformatics methods to predict promoter the sequence of canine BRCA1 gene, then analysis method with promoter activity was verified. The results showed that the prediction of canine BRCA1 promoter reporter gene activity sub group was significantly higher than two of the negative control group (P0.01) for detection. Mutations in the promoter region of BRCA1 canine breast tumor, canine breast tumor tissue samples obtained from the China Agricultural University animal hospital, according to the pathological features of the samples are divided into benign breast tumor group and malignant breast tumor group, each group had 15 cases of tissue samples, and 10 cases of normal canine Sichuan breast tissue as the control group. The PCR products sequencing analysis, mutation of BRCA1 promoter region were tissues. Promoter sequencing results showed that in normal breast tissues and benign breast tumors, were not detected mutations in the promoter region; in malignant breast tumor, found in the promoter region of a 19959098Cdel point mutation, the mutation rate was 46.7% (7/15). In order to detect the expression of BRCA1 in canine mammary tumor changes respectively by the method of quantitative PCR and immunohistochemistry, the expression of BRCA1 gene and mRNA protein was detected in the two groups . the results showed that the dogs in benign and malignant breast tumors, BRCA1 expression of mRNA in the control group were significantly lower than that in normal tissue (P0.05); the expression of BRCA1, malignant breast tumor group was significantly lower than the control group of normal breast tissue (P0.05). The methylation status of BRCA1 encoding region for detection of canine mammary gland tumor mutation and promoter, respectively by PCR sequencing and bisulfite sequencing, measured in the two groups, the methylation status of BRCA1 encoding region mutation and promoter region. The results showed that in normal breast tissues, BRCA1 was not detected in the promoter region mutation; in breast cancer, detected 4 mutations of BRCA1 encoding region, of which 3 was a new mutation. In normal breast tissues and benign breast tumor tissue samples of DNA, were not detected by BRCA1 The promoter methylation in DNA samples; 1 cases of malignant breast tumor tissues, to detect BRCA1 methylation in promoter region. To sum up, in canine malignant mammary tumors, detected a BRCA1 located in the promoter region of the high mutation rate of point mutation. In benign and malignant canine breast tumor BRCA1, the level of mRNA was down regulated. In canine malignant breast tumors, BRCA1 protein level was down regulated. In canine breast tumor detection, 3 new points are located in the encoding region of the mutation. In malignant breast tumors in dogs, to detect BRCA1 methylation in promoter region.
