tdTomato轉(zhuǎn)基因小鼠的建系及其在細(xì)胞示蹤方面的應(yīng)用研究

目的:1.構(gòu)建穩(wěn)定表達(dá)的tandem-dimer Tomato(tdTomato)轉(zhuǎn)基因小鼠并建系,觀察其全身多器官組織的熒光表達(dá)情況,檢測其生理生化指標(biāo)有無異常,確保其符合實(shí)驗(yàn)小鼠的標(biāo)準(zhǔn);2.提取tdTomato轉(zhuǎn)基因小鼠的神經(jīng)干細(xì)胞,觀察其在活體細(xì)胞移植示蹤實(shí)驗(yàn)中的熒光表達(dá)情況。初步嘗試雙色異體干細(xì)胞共培養(yǎng)實(shí)驗(yàn),意在探究離體細(xì)胞培養(yǎng)以及分化時(shí)熒光表達(dá)的穩(wěn)定性。方法:本實(shí)驗(yàn)分為兩個(gè)部分。第一部分,使用Gateway clone技術(shù)構(gòu)建tdTomato載體后,將過表達(dá)載體通過DNA顯微注射法注入受精卵,然后移植至假孕母鼠體內(nèi),待其發(fā)育足月后自然生產(chǎn)。使用雙向鑒定法鑒定并挑取陽性tdTomato轉(zhuǎn)基因小鼠,置于山西醫(yī)科大學(xué)實(shí)驗(yàn)動(dòng)物中心的屏障環(huán)境中進(jìn)行飼養(yǎng),交叉配種繁殖并建系。送檢生理生化指標(biāo)檢測,與正常野生小鼠進(jìn)行統(tǒng)計(jì)學(xué)分析。第二部分,提取tdTomato轉(zhuǎn)基因小鼠的神經(jīng)干細(xì)胞培養(yǎng)至3代,隨后移植到脊髓損傷小鼠模型的脊髓損傷部位進(jìn)行示蹤觀察。另提取eGFP轉(zhuǎn)基因小鼠的骨髓間充質(zhì)干細(xì)胞和神經(jīng)干細(xì)胞分別與tdTomato轉(zhuǎn)基因小鼠的神經(jīng)干細(xì)胞進(jìn)行雙色異體干細(xì)胞共培養(yǎng)實(shí)驗(yàn),跟蹤拍攝干細(xì)胞分化情...
