棕櫚酸促進(jìn)IP3R1蛋白穩(wěn)定性誘導(dǎo)肝細(xì)胞線粒體功能障礙的機(jī)制探索

目的:代謝相關(guān)脂肪性肝病(MAFLD),既往又稱非酒精性脂肪性肝。∟AFLD),是代謝綜合征的肝臟表型,是以肝細(xì)胞脂肪變性和脂質(zhì)沉積為主要特征的一種慢性代謝性疾病。世界范圍內(nèi)NAFLD的人群患病率約為25.24%,最新流行病學(xué)結(jié)果顯示:2019年我國NAFLD患病率約29.8%。NAFLD的臨床病理學(xué)范圍包括非酒精性脂肪肝(NAFL)或單純性脂肪肝,非酒精性脂肪性肝炎(NASH),肝纖維化和肝硬化。棕櫚酸(PA)誘導(dǎo)的肝細(xì)胞凋亡在NAFLD的進(jìn)展中至關(guān)重要。1,4,5-三磷酸肌醇1型受體(IP3R1)是細(xì)胞內(nèi)Ca2+釋放通道,參與PA誘導(dǎo)的肝細(xì)胞凋亡。盡管NAFLD患者和PA處理的肝細(xì)胞中IP3R1蛋白的表達(dá)都表現(xiàn)為明顯升高,但目前尚不清楚PA如何促進(jìn)IP3R1的表達(dá)。因此,本研究擬探索PA處理肝細(xì)胞后IP3R1蛋白表達(dá)增加的原因,并初步探討其分子機(jī)制。方法:第一部分:不同濃度PA處理肝細(xì)胞,進(jìn)行檢測(cè)線粒體膜電位、凋亡蛋白cleaved-caspase3和細(xì)胞凋亡,以確定誘導(dǎo)肝細(xì)胞線粒體功能障礙和細(xì)胞凋亡的最適PA濃度,并用MitoSOX染色檢測(cè)了該最適PA濃度...
