兩種保健食品原料來源中藥的化學(xué)成分研究

【摘要】:本課題對兩種具有保健功能作用的中藥-蒙古黃茜(Astragalus membranaceus(Fisch.)Bge.Var.Mongholicus(Bge.)Hisao)和梔子花(the flower of Gardenia jasmino idesEllis)的化學(xué)成分及初步的藥理活性進(jìn)行了研究。對蒙古黃芪根莖的70%乙醇提取物進(jìn)行了較系統(tǒng)的化學(xué)成分分離及鑒定,從中得到十四個(gè)化合物,包括五個(gè)環(huán)菠蘿蜜烷型三萜皂苷,即Astragaloside Ⅰ(1)、Astragaloside Ⅱ(2)、Isoastragaloside Ⅱ(3)、Isoastragaloside Ⅰ(4)、Astragal oside Ⅳ(5),兩個(gè)羽扇豆烷型三萜羽扇豆酮(6)、白樺脂醇(7),四個(gè)黃酮類化合物(±)-10-甲氧基美迪紫檀素(8)、Isomucronulatol7-glueosid(9)、7,3 '-diohydroxy-5 '-methoxisofiavone(10)、毛蕊異黃酮苷(12),兩個(gè)甾體類化合物β-谷甾醇(13)、胡蘿卜苷(14),一個(gè)酰胺類化合物(2S,3S,4R,10E)-2-[(2R)-2-羥基二十四烷酰氨基]-10-十八烷-1,3,4-三醇(11)。從中可看出Cycloartane型三萜為其主要成分。對梔子花的95%乙醇提取物乙酸乙酯萃取部位分離并鑒定了 11個(gè)化合物,包括九個(gè)Cycloartane型三萜類化合物以及兩個(gè)環(huán)烯醚萜類化合物,其中三個(gè)為新的 Cycloartane 型裂環(huán)三萜,即 Dikamaliartane G(15),Dikamaliartane H(16),DikamaliartaneⅠ(17),一個(gè)已知的 Cycloartane型裂環(huán)三萜 DikamaliartaneD(18),兩個(gè)已知的Cycloartane型三萜梔子花甲酸(19)和梔子花乙酸(20),三個(gè)已知的五環(huán)三萜熊果酸(21)、鐵冬青酸(22)、3β-6β-23-trihydroxyolean-12-en-28-oic acid(23),以及兩個(gè)已知的環(huán)烯醚萜Aglucone(24)和Cyclopenta(25)。對上述化合物通過多種波譜技術(shù)(核磁、質(zhì)譜、紅外等)進(jìn)行了結(jié)構(gòu)鑒定,并對這兩味中藥中的特征成分-Cycloartane型三萜進(jìn)行的體外抗肝損傷藥理活性研究,發(fā)現(xiàn)一些化合物表現(xiàn)出對H2O2誘導(dǎo)的HepG2細(xì)胞損傷的保護(hù)活性。本文的研究工作為進(jìn)一步闡明這兩種中藥保健功能的物質(zhì)基礎(chǔ)提供了重要參考依據(jù)。
