Toll樣受體mRNA和相關(guān)因子在肝泡型包蟲病人體內(nèi)的變化

【摘要】:目的:探討Toll樣受體2(Toll-like receptor 2,TLR2)和TLR4 m RNA在肝多房棘球蚴病(hepatic alveolar echinococcosis,HAE)患者外周血單個(gè)核細(xì)胞(peripheral blood mononuclear cells,PBMC)和肝組織中的表達(dá)情況及其與血漿相關(guān)細(xì)胞因子水平之間的相關(guān)性。方法:收集肝多房棘球蚴病患者(HAE組)28例和健康志愿者作為對(duì)照(HC組,n=28例),用ELISA檢測(cè)血漿中γ干擾素(IFN-γ)、白細(xì)胞介素5(IL-5)、IL-23、IL-10水平,用q RT-PCR法檢測(cè)PBMCs以及HAE患者肝組織中TLR2和TLR4m RNA的表達(dá)水平,外周血嗜酸粒細(xì)胞百分比(Eo%)用血細(xì)胞分析儀檢測(cè)。Spearman秩相關(guān)分PBMCs中TLR2和TLR4 m RNA水平與血漿中細(xì)胞因子水平和外周血Eo%的相關(guān)性。結(jié)果:ELISA檢測(cè)結(jié)果顯示,HAE組血漿中IFN-γ、IL-5、IL-23、IL-10水平分別為(301.100±47.290)、(43.420±11.380)、(86.580±31.990)、(8.766±7.568)pg/ml,均高于HC組的(301.100±67.790)、(40.970±6.310)、(46.770±15.490)、(6.272±10.360)pg/ml,其中兩組的IL-23水平之間的差異性有統(tǒng)計(jì)學(xué)意義(P0.01)。q RT-PCR檢測(cè)結(jié)果顯示,HAE組PBMCs中TLR2 m RNA相對(duì)表達(dá)量為0.100±0.084,高于HC組的0.055±0.040(P0.05);HAE組和HC組的TLR4 m RNA相對(duì)表達(dá)量分別為0.004±0.003和0.003±0.002,兩者間差異無統(tǒng)計(jì)學(xué)意義(P0.05)。HAE患者肝病灶組織中TLR2和TLR4 m RNA相對(duì)表達(dá)量分別為29.680±25.650和21.340±16.640,均高于病灶旁的2.308±4.140和5.541±9.233以及正常組織的1.112±1.431和1.100±1.734(P0.01)。HAE組和HC組外周血Eo%分別為0.448±0.240和0.110±0.100,兩者差異有統(tǒng)計(jì)學(xué)意義P0.01)。Spearman秩相關(guān)分析結(jié)果顯示,PBMCs中TLR2 m RNA水平與血漿中IL-23水平和外周血Eo%之間均存在正相關(guān)關(guān)系(r分別為0.368和0.382)。結(jié)論:肝多房棘球蚴病患者PBMCs、肝病灶中的TLR2和TLR 4m RNA表達(dá)水平升高,PBMCs中TLR2 m RNA水平與血漿中IL-23水平和外周血Eo%之間均存在正相關(guān)關(guān)系。這與棘球蚴絳蟲在感染人體干預(yù)宿主的免疫調(diào)節(jié)系統(tǒng)并產(chǎn)生免疫耐受、免疫逃避和致病有關(guān)。
[Abstract]:Objective: to investigate the expression of Toll like receptor 2(Toll-like receptor 2 (TLR2) and TLR4 m RNA in peripheral blood mononuclear cells (PBMC) and liver tissues of patients with hepatic alveolar echinococcus coensis (HAE) and their correlation with plasma cytokines levels. Methods: the plasma levels of IFN- 緯 and IL-23 IL-10 were measured by ELISA in 28 patients with hepatic hydatidosis and 28 healthy volunteers as control group. The expression of TLR2 and TLR4m RNA in the liver tissues of PBMCs and HAE patients was detected by Q RT-PCR method. The correlation between the levels of TLR2 and TLR4 m RNA in PBMCs and the levels of cytokines in plasma and Eo% in peripheral blood were detected by blood cell analyzer. 緇撴灉:ELISA媯,
