【摘要】：目的:觀察CXCL13對(duì)RANKL在骨性關(guān)節(jié)炎滑膜細(xì)胞中的表達(dá)的影響,探討兩者在骨性關(guān)節(jié)炎滑膜細(xì)胞中的相關(guān)性。方法:第一部分實(shí)驗(yàn)利用TNF-α處理正常關(guān)節(jié)滑膜細(xì)胞,應(yīng)用免疫熒光檢測(cè)滑膜細(xì)胞中CXCL13的表達(dá);第二部分實(shí)驗(yàn)使用CXCL13處理骨性關(guān)節(jié)炎滑膜細(xì)胞,運(yùn)用蛋白免疫印跡分析方法檢測(cè)經(jīng)過(guò)CXCL13(不同濃度、不同時(shí)間)處理后的骨性關(guān)節(jié)炎滑膜細(xì)胞中RANKL蛋白的表達(dá)。結(jié)果:1.免疫熒光結(jié)果顯示CXCL13在骨性關(guān)節(jié)炎滑膜細(xì)胞中表達(dá)明顯低于正常人的滑膜細(xì)胞。2.利用TNF-α(濃度10ug/L)處理正常關(guān)節(jié)滑膜細(xì)胞,隨著作用時(shí)間的延長(zhǎng)CXC113表達(dá)呈現(xiàn)減弱趨勢(shì)。3.蛋白免疫印記檢測(cè)結(jié)果顯示:在骨性關(guān)節(jié)炎滑膜細(xì)胞中RANKL蛋白的表達(dá)量明顯高于正常人滑膜細(xì)胞;經(jīng)TNF-α(濃度為10ng/ml)處理24h后,更換含有CXCL13(濃度25ng/ml)的培養(yǎng)液繼續(xù)培養(yǎng)1h、3h、6h、24h后,發(fā)現(xiàn)在骨性關(guān)節(jié)炎滑膜細(xì)胞中RANKL蛋白表達(dá)水平隨著時(shí)間的延長(zhǎng)而降低。經(jīng)TNF-α(濃度10ng/ml)處理24h后,在更換含有不同濃度的趨化因子CXCL13(Ong/ml、5ng/ml、15ng/ml、25ng/ml)培養(yǎng)液繼續(xù)處理24h后,RANKL蛋白的表達(dá)量隨著濃度的增加而降低。在骨性關(guān)節(jié)炎滑膜細(xì)胞中RANKL的蛋白表達(dá)量隨著CXC113刺激濃度的增加及作用時(shí)間的延長(zhǎng),呈現(xiàn)時(shí)間依賴性及濃度依賴性降低。結(jié)論:CXCL13在一定濃度和時(shí)間能夠?qū)ANKL表達(dá)進(jìn)行抑制,從而可能通過(guò)這一途徑進(jìn)一步延緩骨性關(guān)節(jié)炎癥的炎癥進(jìn)展、骨質(zhì)破壞、軟骨硬化等癥狀。
[Abstract]:Aim: to observe the effect of CXCL13 on the expression of RANKL in synovial cells of osteoarthritis and to explore the correlation between RANKL and RANKL in synovial cells of osteoarthritis. Methods: normal synovial cells were treated with TNF- 偽 and the expression of CXCL13 in synovial cells was detected by immunofluorescence. In the second part, CXCL13 was used to treat synovial cells of osteoarthritis. The expression of RANKL protein in synovial cells of osteoarthritis treated with CXCL13 (different concentrations and different time) was detected by Western blot. The result is 1: 1. Immunofluorescence results showed that the expression of CXCL13 in synovial cells of osteoarthritis was significantly lower than that in normal synovial cells. Normal synovial cells were treated with TNF- 偽 (10 ug- / L), and the expression of CXC113 decreased with the prolongation of time. The expression of RANKL protein in synovial cells of osteoarthritis was significantly higher than that in normal synovial cells, and after 24 hours of TNF- 偽 (10 ng / ml) treatment, the culture medium containing CXCL13 (25 ng / ml) was continuously cultured for 1 h and 3 h for 6 h. It was found that the expression of RANKL protein decreased with time in synovial cells of osteoarthritis. After being treated with TNF- 偽 (10 ng / ml) for 24 h, the expression of RANKL protein decreased with the increase of concentration of CXCL13 Ong / ml 5 ng / ml ~ (15) ng / ml ~ (25) ng 路ml ~ (-1) after 24 h treatment with TNF- 偽 (concentration 10 ng / ml). The expression of RANKL protein in synovial cells of osteoarthritis decreased in a time-and concentration-dependent manner with the increase of CXC113 stimulation concentration and the prolongation of the time of action. Conclusion the expression of RANKL can be inhibited at a certain concentration and time, which may further delay the progression of inflammation, bone destruction and cartilage sclerosis in osteoarthritis.
