藏醫(yī)名方聯(lián)用對(duì)腦缺血損傷及行為學(xué)障礙的藥效學(xué)研究

【摘要】:研究發(fā)現(xiàn),民族藥物在針對(duì)腦卒中和相關(guān)后遺癥的臨床治療方面依然保持有高漲之勢(shì),其中以藏藥最為突出。在經(jīng)典藏醫(yī)醫(yī)著中記載到,對(duì)腦卒中病癥都以血、脈、隆三者結(jié)合的方法,達(dá)到標(biāo)本同治的目的。綜合來(lái)看,藏醫(yī)積累的經(jīng)驗(yàn)及其獨(dú)特的時(shí)辰給藥理論都是治療腦缺血類(lèi)疾病的優(yōu)勢(shì)所在。本課題基于以上特點(diǎn)探索三種藏醫(yī)名方二十五味珊瑚丸、如意珍寶丸、二十味沉香丸于早、中、晚聯(lián)用對(duì)腦缺血損傷及行為學(xué)障礙的影響。1.建立MCAO/R模型,腦組織Nissl染色,觀(guān)察神經(jīng)元Nissl小體變化表明:藏醫(yī)名方聯(lián)用可有效減輕腦缺血受損引起的Nissl小體損傷,維護(hù)神經(jīng)元結(jié)構(gòu)完整性。采用IHC/S-P法和RT-PCR定量結(jié)果表明:藏醫(yī)名方聯(lián)用能有效促進(jìn)Bcl-2蛋白含量,下調(diào)Caspase-3、NF-κBp65、ICAM-1、VEGF、TLR4蛋白表達(dá),對(duì)腦缺血損傷引起的炎癥反應(yīng)及細(xì)胞凋亡有較好的改善作用。2.建立MCAO/R模型,血液流變學(xué)指標(biāo)檢測(cè)結(jié)果表明:藏醫(yī)名方聯(lián)用可顯著降低缺血性腦卒中血管內(nèi)全血粘度,血漿粘度,ESR,血沉方程K值,Hct,EAI,紅細(xì)胞剛性指數(shù);顯著升高電泳指數(shù),紅細(xì)胞變形指數(shù),改善血液流動(dòng)力學(xué)特性;建立小鼠急性血瘀模型,耳廓微循環(huán)檢測(cè)表明:藏醫(yī)名方聯(lián)用顯著擴(kuò)張腎上腺素所致急性血瘀模型小鼠耳廓微動(dòng)脈、微靜脈血管管徑,加快血液流速,改善微循環(huán)。結(jié)合MCAO/R模型和小鼠急性血於模型,共同探討三種藏醫(yī)名方活血化瘀、擴(kuò)張血管、促進(jìn)血液流動(dòng)及對(duì)血管功能的保護(hù)。3.建立小鼠記憶獲得障礙模型,利用小鼠趨暗習(xí)性對(duì)其學(xué)習(xí)、記憶、反應(yīng)能力測(cè)試結(jié)果表明:藏醫(yī)名聯(lián)用顯著增加小鼠潛伏期、主動(dòng)穿梭次數(shù),且測(cè)試成績(jī)優(yōu)于訓(xùn)練成績(jī),保護(hù)M受體抑制劑所致小鼠腦損傷,減輕小鼠產(chǎn)生的學(xué)習(xí)記憶反應(yīng)障礙的癥狀;有效降低小鼠腦組織中AChE、MDA活性,顯著升高SOD含量,維護(hù)膽堿能神經(jīng)功能,清除體內(nèi)自由基,降低脂質(zhì)過(guò)氧化損傷。
[Abstract]:The study found that there is still an upsurge in the clinical treatment of stroke and related sequelae, especially Tibetan medicine. In classical Tibetan medicine, it is recorded that blood and veins are used to treat stroke. The method of combination of the three Longs, to achieve the purpose of the same treatment of specimens. In a comprehensive view, The accumulated experience of Tibetan medicine and its unique theory of time administration are the advantages of treating cerebral ischemia diseases. Based on the above characteristics, this paper explores three kinds of famous Tibetan medicine prescription 25 coral pills, Ruyi Zhenbao pills, 20 kinds of Shenxiang pills in the early and middle. The effects of late combined use on cerebral ischemia injury and behavioral disorders. 1. MCAO/R model was established, brain tissue Nissl staining and changes of neuron Nissl corpuscles were observed. The results showed that Tibetan medicine combined with famous prescription could effectively reduce Nissl corpuscle injury caused by cerebral ischemia injury. The results of IHC/S-P and RT-PCR quantitative analysis showed that the combination of Tibetan medicine and famous prescriptions could effectively promote the content of Bcl-2 protein and down-regulate the expression of Caspase-3, NF- 魏 Bp65, ICAM-1 and VEGFTLR4 protein. The model of MCAO/R was established. The results of hemorheological index test showed that the combination of Tibetan medicine and famous prescription could significantly reduce the whole blood viscosity of blood vessel in ischemic stroke. Plasma viscosity, ESR, erythrocyte rigidity index, erythrocyte rigidity index, erythrocyte deformability index, erythrocyte deformability index, erythrocyte deformability index, and establishment of acute blood stasis model in mice. The detection of auricle microcirculation showed that the acute blood stasis model of mice induced by epinephrine was significantly expanded by the famous prescription of Tibetan medicine, which could accelerate the blood flow rate and improve the microcirculation, combining with the MCAO/R model and the acute blood flow model of mice, the diameter of the arterioles and venules of the auricle of the mice was increased. To explore three famous Tibetan medicine prescriptions, such as activating blood circulation and removing blood stasis, dilating blood vessels, promoting blood flow and protecting vascular function .3.To establish a model of memory acquisition disorder in mice, and to learn and remember it by using dark habit of mice. The results of response ability test showed that the combination of Tibetan medicine and Chinese medicine could significantly increase the latency and the number of active shuttles in mice, and the test scores were better than the training results, which protected mice from brain injury induced by M receptor inhibitors. The symptoms of learning and memory reaction in mice were alleviated, the activity of ache MDA in brain tissue was decreased, the content of SOD was increased significantly, cholinergic nerve function was maintained, free radical was cleared and lipid peroxidation injury was reduced.
