【摘要】:目的:我們擬尋找與無(wú)功能型垂體腺瘤的侵襲性相關(guān)的重要蛋白,為全面、系統(tǒng)研究無(wú)功能型垂體腺瘤侵襲生長(zhǎng)的機(jī)制提供理論依據(jù),為今后利用蛋白質(zhì)組學(xué)方法全面、系統(tǒng)研究無(wú)功能型垂體腺瘤的病因,以及分子靶向治療、基因治療提供新的突破點(diǎn)。方法:收集2015年10月至2016年10月期間新疆維吾爾自治區(qū)人民醫(yī)院神經(jīng)外科收治的經(jīng)本院手術(shù)治療的18例無(wú)功能型垂體瘤病人的腫瘤標(biāo)本(侵襲性生長(zhǎng)9例,非侵襲性生長(zhǎng)9例)。用二維凝膠電泳結(jié)合質(zhì)譜技術(shù)尋找在無(wú)功能型垂體腺瘤中表達(dá)的差異性蛋白;應(yīng)用雙向凝膠電泳(2-DE)結(jié)合基質(zhì)輔助激光解析電離飛行時(shí)間質(zhì)譜技術(shù)(MALDI-TOF-MS)對(duì)侵襲性無(wú)功能型垂體腺瘤及非侵襲性無(wú)功能型垂體腺瘤腫瘤組織進(jìn)行蛋白質(zhì)翻譯水平的篩選,并比較組織蛋白的異同,尋找與無(wú)功能型垂體腺瘤侵襲性相關(guān)的蛋白質(zhì)及可能在臨床診斷中應(yīng)用的蛋白質(zhì)。結(jié)果:(1)通過(guò)雙向電泳結(jié)果(通過(guò)PDQuest軟件顯不同組的差異表達(dá)蛋白質(zhì)點(diǎn)),建立了無(wú)功能型垂體腺瘤的的2-DE圖譜。(2)通過(guò)研究我們找到4種差異蛋白點(diǎn),其中p14ARF蛋白在侵襲性無(wú)功能型垂體腺瘤的腫瘤組織中較非侵襲性無(wú)功能型垂體腺瘤的腫瘤表達(dá)下調(diào),EGFR、HMGA2、PIK3CA蛋白在侵襲性無(wú)功能型垂體腺瘤的腫瘤組織中較非侵襲性無(wú)功能型垂體腺瘤的腫瘤表達(dá)上調(diào)。結(jié)論:這4種差異蛋白可能與無(wú)功能型垂體腺瘤的侵襲性生長(zhǎng)有關(guān)。
[Abstract]:Objective: to search for important proteins related to invasiveness of non-functional pituitary adenomas, and to provide theoretical basis for a comprehensive and systematic study of the mechanism of invasiveness and growth of non-functional pituitary adenomas. In order to study the etiology and molecular targeted therapy of non-functional pituitary adenoma systematically by proteomics. Gene therapy provides a new breakthrough. Methods:. From October 2015 to October 2016, 18 patients with disfunctional pituitary adenoma treated in neurosurgery department of Xinjiang Uygur Autonomous region people's Hospital were collected (. Invasive growth was found in 9 cases. Two dimensional gel electrophoresis and mass spectrometry were used to search for differentially expressed proteins in non-functional pituitary adenomas. Two-dimensional gel electrophoresis (2-DE) combined with matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MSM). The protein translation level of invasive and non-invasive pituitary adenomas was screened. The differences and similarities of histone were compared. Search for proteins associated with invasiveness of nonfunctional pituitary adenomas and proteins that may be used in clinical diagnosis. PDQuest software was used to show the differentially expressed protein spots in different groups. The 2-DE map of non-functional pituitary adenoma was established. The expression of p14ARF protein was down-regulated in tumor tissues of invasive non-functional pituitary adenomas than that of non-invasive pituitary adenomas. The expression of PIK3CA protein was up-regulated in invasive non-functional pituitary adenomas than in non-invasive pituitary adenomas. Conclusion:. These four differentially expressed proteins may be related to the invasive growth of nonfunctional pituitary adenomas.
