BTBD7及E-cadherin在唾液腺腺樣囊性癌中的表達與相關(guān)性的研究

【摘要】:目的:檢測BTBD7和E-cadherin在唾液腺腺樣囊性癌(SACC)中的表達情況,并研究其與SACC不同臨床病理特性的相關(guān)關(guān)系,同時研究二者之間的相關(guān)關(guān)系。材料和方法:應(yīng)用免疫組織化學(xué)的方式,將本研究搜集的50例SACC組織標本及10例正常唾液腺組織標本進行染色處理,分別檢測其中BTBD7及E-cadherin的表達情況。抗BTBD7抗體結(jié)合位置為細胞漿,抗E-cadherin抗體結(jié)合位置除細胞漿外,同時有細胞膜;細胞質(zhì)內(nèi)出現(xiàn)大量的棕黃色顆粒狀物質(zhì)著色判定為BTBD7陽性表達的標準,細胞質(zhì)及細胞膜表面同時出現(xiàn)明顯的棕黃色顆粒狀物質(zhì)顯色為E-cadherin陽性表達的標準。用統(tǒng)計學(xué)軟件SPSS 23.0分析實驗結(jié)果,將50例組織標本按照各臨床病理特征(病理類型、神經(jīng)侵犯、血行轉(zhuǎn)移、臨床分期)進行分組,采取x2檢驗和Fisher確切概率法檢驗差異,采取Spearman相關(guān)分析檢驗BTBD7及E-cadherin的關(guān)聯(lián)性;結(jié)果以P0.05標記為存在統(tǒng)計學(xué)差異。結(jié)果:(1)與正常唾液腺組織比較,在唾液腺腺樣囊性癌組織中BTBD7表達顯著增加,在50例腫瘤組織中有21例表達呈陽性,其陽性表達率為420%,兩者之間具有統(tǒng)計學(xué)差異(P=0.011)。BTBD7在低分化SACC中表達增加;在Ⅰ期、Ⅱ期與Ⅲ期、Ⅳ期之間具有統(tǒng)計學(xué)差異(P=0.002)。BTBD7在有神經(jīng)浸潤的SACC組織中陽性表達率高達63.640%,明顯高于無神經(jīng)浸潤的腫瘤組織(21.430%),其差異具有顯著性(P=0.002)。在16例有血行性轉(zhuǎn)移的唾液腺腺樣囊性癌組織中BTBD7的陽性表達率為62.5%,顯著高于無轉(zhuǎn)移的腫瘤組織,有統(tǒng)計學(xué)意義(P=0.022)。(2)與正常唾液腺組織比較,在SACC組織中E-cadherin表達下降(62%),其差異具有統(tǒng)計學(xué)意義(P=0.018)。E-cadherin在低分化SACC中表達減少,在Ⅰ期、Ⅱ期與Ⅲ期,Ⅳ期之間的差異具有統(tǒng)計學(xué)意義(P=0.002)。E-cadherin在有神經(jīng)浸潤的SACC組織中陽性表達率為45.45%,明顯低于無神經(jīng)浸潤的癌組織(78.57%),其差異有統(tǒng)計學(xué)意義(P=0.015)。在有血行性轉(zhuǎn)移的SACC組織中E-cadherin的陽性表達率為37.50%,低于無轉(zhuǎn)移的癌組織(75.86%),差異具有顯著性(P=0.011)。(3)在50例SACC樣本中,有8例表現(xiàn)為BTBD7與E-cadherin共同陽性,6例為共同陰性,二者單獨陽性的例數(shù)分別有23例和6例,經(jīng)過Spearman相關(guān)分析檢驗,二者的表達在SACC中呈現(xiàn)負相關(guān)(r=-0.419,P=0.002)。結(jié)論:BTBD7在SACC中呈現(xiàn)高表達,E-cadherin呈現(xiàn)低表達,且二者與SACC的病理分期、有無神經(jīng)浸潤及有無遠處轉(zhuǎn)移存在相關(guān)性;同時BTBD7和E-cadherin在SACC組織中的表達呈現(xiàn)負相關(guān)。意義:本課題研究了 BTBD7與E-cadherin在唾液腺腺樣囊性癌中的表達情況,結(jié)果發(fā)現(xiàn)二者與SACC的病理分期、神經(jīng)侵犯、遠處轉(zhuǎn)移均相關(guān),同時二者在SACC組織中表達呈現(xiàn)負相關(guān),同時檢測兩者也許可為研究SACC細胞的生物學(xué)行為提供參考指標,為臨床治療SACC提供新思路。
[Abstract]:Objective: to detect the expression of BTBD7 and E-cadherin in salivary adenoid cystic carcinoma (SCC) and to study the relationship between them and the different clinicopathological characteristics of SACC. Materials and methods: 50 SACC tissues and 10 normal salivary gland tissues collected in this study were stained by immunohistochemistry. The expression of BTBD7 and E-cadherin were detected respectively. The binding position of anti-#en1# antibody was cytoplasm, and the binding position of anti-E-cadherin antibody was except cytoplasm. There is cell membrane at the same time; A large number of brown-yellow granules in the cytoplasm were identified as positive expression of BTBD7. The standard of E-cadherin positive expression was observed on the surface of cytoplasm and cell membrane. The results were analyzed by statistical software SPSS 23.0. According to the clinicopathological features (pathological type, nerve invasion, blood metastasis, clinical stage), 50 tissue specimens were divided into two groups. The difference was detected by x2 test and Fisher exact probability method. The correlation between BTBD7 and E-cadherin was examined by Spearman correlation analysis. Results compared with normal salivary gland tissue, the expression of BTBD7 in salivary adenoid cystic carcinoma was significantly increased. Of the 50 cases, 21 were positive, and the positive rate was 420%. There was a significant difference between the two groups in the expression of PTEP 0.011. BTBD7 in poorly differentiated SACC.
