紫蘇籽油及原花青素的提取工藝研究

【摘要】:紫蘇是我國傳統(tǒng)的中藥材和油料作物,紫蘇籽含油量高,皮中含有原花青素,對紫蘇籽進行綜合利用市場前景廣闊。本文以紫蘇籽為原料,采用超聲波輔助水酶法提取紫蘇籽油,優(yōu)化工藝,并測定紫蘇籽油的性質(zhì);同時研究了紫蘇籽皮原花青素的提取及純化工藝,并對其抗氧化活性進行了測定。主要研究結(jié)果如下:(1)在單因素試驗基礎(chǔ)上,通過響應(yīng)面法得到超聲波輔助水酶法提取紫蘇籽油的最優(yōu)條件:復合酶配比(纖維素酶用量5.5%、中性蛋白酶用量4.5%、果膠酶用量7.5%),液料比6:1(m L/g),超聲時間23min,酶解溫度50℃,酶解時間2.5h,出油率為30.45%。(2)對水酶法、熱榨法、冷榨法及有機溶劑浸提法所提取的紫蘇籽油的主要理化性質(zhì)、脂肪酸組成、生物活性成分含量以及抗氧化能力進行了測定與比較,結(jié)果表明:水酶法油的過氧化值最低,碘值最高;水酶法油的棕櫚酸含量低,油酸含量顯著高于熱榨油與有機溶劑浸提油,α-亞麻酸與不飽和脂肪酸含量最高;水酶法油的VE含量與有機溶劑浸提油相差不大,多酚、總黃酮及甾醇含量最高;通過DPPH法、ABTS法和FRAP法得出水酶法油的抗氧化能力最強。綜合來看,水酶法油的品質(zhì)較好。(3)通過單因素和響應(yīng)面法試驗,得到超聲波輔助法提取紫蘇籽皮原花青素的最佳條件為:液料比15:1(m L/g),乙醇體積分數(shù)62%,超聲時間30min,超聲溫度53℃,提取量為2.170mg/g。(4)通過靜態(tài)吸附與解吸附試驗,選擇XDA-8樹脂來純化紫蘇籽皮原花青素,經(jīng)動態(tài)吸附與解吸附試驗得出純化的最優(yōu)條件為:上樣濃度4mg/mL,上樣流速4BV/h,乙醇體積分數(shù)70%,洗脫流速2BV/h,洗脫劑的用量為13BV。純化前的紫蘇籽皮原花青素含量為5.25%,純化后為12.10%,提高了130.48%。對純化前后的紫蘇籽皮原花青素、VC以及BHT四種樣品的抗氧化能力進行測定與比較,發(fā)現(xiàn)純化后的紫蘇籽皮原花青素的抗氧化能力最強。

Perilla seed has a high oil content and contains procyanidins in the skin, so the comprehensive utilization of perilla seed has a broad market prospect.
