土壤放線菌XN-04代謝產(chǎn)物除草活性成份分離純化與結(jié)構(gòu)解析

【摘要】:通過活性篩選,得到土壤放線菌XN-04,其發(fā)酵的次級(jí)代謝產(chǎn)物經(jīng)過生物測(cè)定具有良好的除草活性。本文研究了其具有抑制雜草發(fā)芽生長(zhǎng)活性的次級(jí)代謝產(chǎn)物,并通過相關(guān)手段對(duì)其進(jìn)行了菌種鑒定。對(duì)該菌株XN-04的發(fā)酵次級(jí)代謝產(chǎn)物依次進(jìn)行溶劑萃取、硅膠柱層析、薄層層析、高效液相制備等分離提純手段,分離得到了兩個(gè)具有除草活性的化合物XN-04-A4和XN-04-B2。兩個(gè)化合物經(jīng)過質(zhì)譜(MS)、核磁共振(NMR)等常規(guī)的波譜測(cè)試,最終解析出兩個(gè)化合物的分子式分別是C15H10O5和C15H23NO4,相關(guān)化學(xué)分子量分別為270和281。綜合各類譜圖分析,確定化合物XN-04-A4與文獻(xiàn)報(bào)道的染料木素(Genistein)結(jié)構(gòu)相同;化合物XN-04-B2與文獻(xiàn)報(bào)道的放線菌酮(Naramycin A)結(jié)構(gòu)相同。通過研究,建立了用高效液相色譜法測(cè)定發(fā)酵液中化合物XN-04-A4和XN-04-B2的發(fā)酵效價(jià),并進(jìn)行了化合物XN-04-A4和XN-04-B2除草譜的試驗(yàn)。通過觀察菌株XN-04的培養(yǎng)特征、形態(tài)特征、生理生化特征,綜合16SrDNA基因序列分析,確定該菌株歸屬于鏈霉菌屬,與Streptomycesgalilaeus同源性相似度為99%。
[Abstract]:Soil actinomycetes XN-04 was obtained by activity screening. The secondary metabolites of its fermentation have good herbicidal activity by bioassay. In this paper, the secondary metabolites which have the ability to inhibit the growth of weeds were studied. The fermentation secondary metabolites of XN-04 were separated and purified by solvent extraction, silica gel column chromatography, thin layer chromatography, high performance liquid phase preparation and so on. Two herbicide compounds, XN-04-A4 and XN-04-B2, were isolated. The two compounds were characterized by conventional spectroscopic measurements such as MS MS, NMR and so on. Finally, the molecular formulas of the two compounds are C15H10O5 and C15H23NO4, the molecular weights of which are 270 and 281, respectively. It was confirmed that the structure of the compound XN-04-A4 was the same as that of genistein reported in the literature. The structure of compound XN-04-B2 is the same as that of Naramycin A reported in the literature. The fermentation titers of compounds XN-04-A4 and XN-04-B2 in fermentation broth were determined by HPLC. The experiment of compound XN-04-A4 and XN-04-B2 weeding spectrum was carried out. The culture characteristics, morphological characteristics, physiological and biochemical characteristics of XN-04 were observed. According to the analysis of 16s rDNA gene sequence, the strain belongs to Streptomyces, and the similarity with Streptomycesgalilaeus is 99%.
