赤蘚糖亞硫酸銨法制備焦糖色素工藝與性質(zhì)研究

【摘要】:焦糖色素是一種人工合成的食用色素,也是目前廣泛使用的一種食品添加劑之一。焦糖色素的食用安全性在食品使用中一直受到關注,其中5-羥甲基糠醛(5-HMF)的含量必須得到控制。本文采用赤蘚糖為美拉德反應糖原料,經(jīng)高溫反應制備一種不含5-HMF的焦糖色素,通過一系列實驗確定了制備赤蘚糖焦糖色素(ECP)的最佳工藝條件,再采用超濾工藝得到不同品質(zhì)的ECP產(chǎn)品。分離和鑒定了赤蘚糖美拉德反應中間產(chǎn)物,同時進一步考察了ECP類黑精大分子結(jié)構(gòu),并對其抗腫瘤和抗氧化活性進行了研究,為ECP的生產(chǎn)和應用提供更重要的依據(jù)。 首先通過二次旋轉(zhuǎn)回歸組合設計摸索了ECP制備最佳工藝條件:保溫溫度為140℃(1.682水平),保溫時間為40min(1.682水平),赤蘚糖液濃度459g/L(1水平),(NH4)2SO3含量(以NH4+計)為4%(0水平),此時色率最大值為18054.43EBC。 其次研究了超濾對ECP品質(zhì)的影響,ECP經(jīng)超濾分級后,截留組分特性差異較為明顯。通過分析發(fā)現(xiàn),各級分子段截留產(chǎn)物粒度分布均勻。100kDa以上分子截留段ECP其色率達到95526.32EBC,紅色指數(shù)和黃色指數(shù)分別為3.59和5.51,其品質(zhì)符合市售焦糖色素標準。 參照GB8817-2001,分別從食品的安全和應用兩方面對赤蘚糖焦糖色素各項理化指標進行了研究,結(jié)果表明ECP不含有5-HMF,固形物含量范圍在30%-50%的ECP樹脂化時問在25h以上,產(chǎn)品穩(wěn)定性好,貨架期長;帶負電荷,適用于軟飲料工業(yè)。 分離和鑒定了赤蘚糖美拉德反應中間產(chǎn)物,通過離子碎片峰的組合,得到分離物的化學結(jié)構(gòu)式,其分子式為C8H10O3,從機理上證明了赤蘚糖美拉德反應不產(chǎn)生5-HMF。 最后通過對ECP10k-50kDa,ECP50kDa-100kDa和ECP100kDa-0.22μm三種樣品進行元素分析、紅外以及熱裂解分析,研究了ECP類黑精的大分子結(jié)構(gòu),并考察了類黑精大分子抗腫瘤和抗氧化活性,結(jié)果表明ECP在熱裂解過程中產(chǎn)生了SO2,鏈狀烷烴和呋喃環(huán)結(jié)構(gòu)。不同倍數(shù)ECP分別培養(yǎng)24h和72h后,ECP1Ok-5OkDa對MDA-MB-231細胞有明顯的抑制效果。ECP50k-100kDa開始誘導細胞癌變,ECP100kDa-0.22μm誘導效果最強。
[Abstract]:Caramel pigment is a synthetic food pigment and one of the widely used food additives. Food safety of caramel pigments has been concerned in food use. The content of 5-HMF must be controlled. In this paper, a caramel pigment without 5-HMF was prepared by high temperature reaction using erythosaccharide as raw material for Maillard reaction. The optimum technological conditions for preparing caramel pigment were determined by a series of experiments. Different quality ECP products were obtained by ultrafiltration process. In this paper, the intermediate products of Maillard reaction of erythrum were isolated and identified, and the macromolecular structure of ECP melanin was further investigated, and its anti-tumor and anti-oxidation activities were studied, which provided a more important basis for the production and application of ECP.
