【摘要】：在引入固相萃取(SPE)C4前處理技術(shù)的基礎(chǔ)上,首次采用氣相色譜質(zhì)譜聯(lián)用(GC-MS)和離子色譜(ICS)技術(shù)定性、定量化評估了SPE C4柱對常見文物保存環(huán)境中可溶鹽離子的排除效果,進一步探討了該技術(shù)對氨基酸分析結(jié)果的影響、保護色譜柱的效果及對文物樣品測試的適用性。結(jié)果表明,SPE C4技術(shù)能有效排除Na~+,K~+,Mg~(2+),Ca~(2+),Cl~-,NO_3~-,SO_4~(2-)等常見可溶鹽離子,平均洗脫率(脫鹽率)達99.99%;添加30 g/L NaCl的豬皮膠經(jīng)SPE C4處理后測得的色譜圖出峰完整,未檢測到Na~+和Cl~-,11種氨基酸含量測量結(jié)果的平均相對誤差約5%。文物樣品氨基酸色譜圖基線平穩(wěn)、分離良好。因此,SPE C4技術(shù)適用于中國古代蛋白膠料氣相色譜分析的前處理中,對分析鑒定我國古代文物蛋白膠料具有極高的實用價值。
[Abstract]:Based on the introduction of solid-phase extraction (SPE)-C _ 4) pretreatment technology, gas chromatography-mass spectrometry (GC-MS) and ion chromatography-(ICS) were used for the first time. The effect of SPE C4 column on the removal of soluble salt ions in the preservation environment of common cultural relics was quantitatively evaluated. The influence of the technique on amino acid analysis results, the effect of protection column and the applicability of the technique to the determination of cultural relics samples were further discussed. The results showed that SPE C4 technology could effectively eliminate the common soluble salt ions such as Na~, K ~, Mg~ (2), Ca~ (2), Cl~-,NO_3~-,SO_4~ (2 -), and the average desalination rate (desalination rate) was 99.99%. After being treated with 30 g / L NaCl, the chromatogram showed a complete peak, and the average relative error of Na~ and Cl~-,11 amino acid content measurement results was about 5%. The baseline of amino acid chromatogram of cultural relics is stable and the separation is good. Therefore, SPE-C4 technology is suitable for the pre-treatment of Chinese ancient protein compound by gas chromatography, and it is of great practical value for the analysis and identification of protein compound of ancient cultural relics in China.
