江陵天星觀一號楚墓出土飽水木漆器F455中細菌對硬松木的腐蝕研究

【摘要】:研究木質(zhì)文物中的細菌對木材腐蝕性影響,篩選防腐劑抑制腐蝕性強細菌的腐蝕作用,是延長木質(zhì)文物保存期的一種有效的生物保護措施。為研究飽水木漆器F455及其水環(huán)境中細菌的種類,以及對木材的腐蝕程度,采用16S rRNA基因序列分析方法及生理生化試驗,對飽水木漆器F455及水環(huán)境中細菌進行鑒定,并選取典型菌按5×10~8個/瓶菌量接種馬尾松心材(懸于無菌自來水中),37℃培養(yǎng)120d,測試木材的損失率。結(jié)果發(fā)現(xiàn),從F455文物挑選的32株細菌中,有29株菌鑒定為芽孢桿菌屬(Bacillus),為優(yōu)勢菌屬,其余為短桿菌屬(Brevibacterium)、短波單胞菌屬(Brevundimonas)和產(chǎn)堿桿菌屬(Alcaligenes)各1株。水樣樣品中細菌菌屬較F455文物樣品中豐富,27株菌有9個屬,其中短桿菌屬(Brevibacterium)有9株,為優(yōu)勢菌屬,其余交替赤細菌屬(Altererythrobacter)有5株,短波單胞菌屬(Brevundimonas)有4株,產(chǎn)堿桿菌屬(Alcaligenes)有3株,黃桿菌屬(Flavobacterium)有2株,芽孢桿菌屬(Bacillus sp.)、白色桿菌屬(Leucobacter)、類芽孢桿菌屬(Paenibacillus)、蒼白桿菌屬(Ochrobactrum)各1株。從2種樣品材料的分離菌中共挑選出14株典型菌進行木塊腐蝕試驗,與對照組比較,其中有9株菌差異極顯著。說明這些菌對馬尾松木材有一定的腐蝕作用,但是腐蝕率非常低,最高僅1.56%。表明這些細菌對試驗木材馬尾松腐蝕并不嚴重。該研究分析了飽水木漆器長期保存在水槽中細菌對木漆器的影響,豐富了飽水木漆器文物生物影響的理論依據(jù)。
