ABO基因分型及其法庭科學(xué)應(yīng)用研究

【摘要】:目的 為使ABO基因型更好地服務(wù)于法庭科學(xué)實踐,本研究建立和優(yōu)化ABO基因分型方法,并充分評估ABO基因型在法庭科學(xué)實踐中的可信性及應(yīng)用價值。方法 采用PCR-RFLP技術(shù),復(fù)合擴增產(chǎn)物用Kpn Ⅰ和Alu Ⅰ限制性內(nèi)切酶酶切,經(jīng)聚丙烯酰胺凝膠電泳及硝酸銀染色判讀ABO基因型。運用該方法對血樣、混合斑、唾液斑、毛發(fā)、肌肉、骨等常見生物學(xué)樣本進行ABO基因分型。用PCR-直接測序法對ABO基因進行序列分析。結(jié)果 該方法將ABO血型直接區(qū)分為常見的六種基因型。兩代家系的ABO基因型結(jié)果均符合孟德爾遺傳規(guī)律。同一個體的ABO基因型與ABO表型結(jié)果完全相符。100例認定嫌疑人的強奸案中混合斑精子的ABO基因型與相應(yīng)嫌疑人血樣的ABO基因型結(jié)果完全一致。常見生物學(xué)檢材均能得到準(zhǔn)確的ABO基因型檢測結(jié)果。經(jīng)PCR-直接測序法檢出一例ABO基因突變。結(jié)論 運用PCR-RFLP
