HPLC法測定甲基苯丙胺與苯丙胺

【摘要】:目的建立甲基苯丙胺(methamphetamine,MA)和苯丙胺(amphetamine,AMP)的反相高效液相色譜測定方法。方法采用C18柱,以甲醇-磷酸鹽緩沖液為流動相,流速1.0mL/min,檢測波長為215nm,同時(shí)收集190~360nm的紫外光譜圖,并以此與保留時(shí)間作為定性依據(jù)。在定性的基礎(chǔ)上,建立定量檢測方法,并對方法進(jìn)行評價(jià)研究。結(jié)果所建方法能良好分離MA和AMP,結(jié)合判斷依據(jù)能準(zhǔn)確定性;MA在1.4~270μg/mL濃度范圍內(nèi)線性關(guān)系良好,R2=1,日內(nèi)標(biāo)準(zhǔn)偏差(RSD)與日間RSD均2.4%,檢出限為0.73μg/mL,平均加樣回收率為102.5%;AMP在0.9~580μg/mL濃度范圍內(nèi)線性關(guān)系良好,R2=0.9999,日內(nèi)RSD與日間RSD均2.3%,檢出限為0.52μg/mL,平均加樣回收率為101.7%。結(jié)論此方法簡便、快速、準(zhǔn)確,適用于MA與AMP的檢測。
[Abstract]:Objective to establish a reversed-phase HPLC method for the determination of methamphetamine (methamphetamine,MA) and amphetamine (amphetamine,AMP). Methods C _ (18) column was used with methanol-phosphate buffer as mobile phase, flow rate was 1.0 mL / min, detection wavelength was 215 nm, UV spectra of 190~360nm were collected, and retention time was used as qualitative basis. On the basis of qualitative analysis, a quantitative detection method was established and evaluated. Results the method can be used to separate MA and AMP, correctly and qualitatively. MA was linear in the range of 1.4 渭 g/mL and 270 渭 g/mL, R2X 1, (RSD) and RSD were 2.4, the detection limit was 0.73 渭 g / mL, and the average recovery was 102.5%. The linear range of AMP was 0.9580渭 g/mL, R2O0.99999.The detection limit of RSD was 0.52 渭 g / mL, and the average recovery was 101.7%, both RSD and RSD were 2.3 渭 g 路mL ~ (-1), 0.52 渭 g 路mL ~ (-1) 路min ~ (-1) 路min ~ (-1). Conclusion this method is simple, rapid, accurate and suitable for the detection of MA and AMP.
