腦震蕩鼠認(rèn)知行為障礙及腦乙酰膽堿轉(zhuǎn)移酶(ChAT)變化的研究

【摘要】:目的:應(yīng)用腦震蕩(Cerebral Concussion,CC)大鼠模型,觀察不同損傷程度腦震蕩后不同時間大鼠認(rèn)知障礙變化及乙酰膽堿轉(zhuǎn)移酶變化規(guī)律,探討腦震蕩鼠認(rèn)知、記憶障礙的特點(diǎn)及與乙酰膽堿轉(zhuǎn)移酶變化的關(guān)系,了解腦震蕩鼠認(rèn)知記憶行為障礙的神經(jīng)生物學(xué)基礎(chǔ),為腦震蕩后綜合征的臨床診斷、治療提供理論依據(jù)。方法:應(yīng)用自制金屬單擺式閉合性腦損傷機(jī)械打擊裝置復(fù)制腦震蕩大鼠模型;70只SD大鼠隨機(jī)分為8組:大鼠單純性腦震蕩組(Pure Cerebral Concussion,PCC)傷后1、2、4、8、16、24天組共六個損傷組(n=10)、復(fù)雜性腦震蕩組(Complicated Cerebral Concussion,CCC)和正常對照組(n=10)。應(yīng)用平衡木(Beam Balance test,BBT)及噪音橫桿跑動試驗(yàn)(Beam Walking test,BWT),分別檢測和傷后1~5天的平衡時間(Balance Latency,BL)以及步入黑箱的潛伏期(Step Through Latency,STL)的改變。應(yīng)用免疫印記法(Western Blot)分別檢測大鼠PCC傷后1、2、4、8、16、24天中隔區(qū)、基底節(jié)、前額葉和中腦的乙酰膽堿轉(zhuǎn)移酶(Cholin Acetyltransferase,CHAT)的變化。結(jié)果:行為學(xué)結(jié)果顯示PCC組平衡時間與對照組比較無差異;但在BWT中,僅在1、2天步入潛伏期顯著延長。CCC組平衡時間與對照組比較,在傷后第1~3天差異具有顯著性(p<0.05);在BWT中,,第1~5天步入潛伏期均顯著延長,逐漸從48.12±18.24秒恢復(fù)至27.10±28.36秒。免疫印跡結(jié)果顯示與對照組相比較,基底節(jié)ChAT傷后第2天開始上升,差異具有顯著性(p<0.05);傷后第24天表達(dá)有所下降但仍未恢復(fù)至正常。中隔區(qū)ChAT傷后第1天開始上升,但差異不具有顯著性(p>0.05);直至傷后第4~24天差異均具有顯著性(p<0.05)。前額葉ChAT傷后第1天開始下降,但差異不具有顯著性(p>0.05);直至傷后第8~24天差異均具有顯著性(p<0.05)。中腦ChAT傷后第2天開始上升,差異具有顯著性(p<0.05);直至傷后第24天差異均具有顯著性(p<0.05)。結(jié)論:1、腦震蕩后大鼠在BBT和BWT實(shí)驗(yàn)中出現(xiàn)程度不同的近期學(xué)習(xí)記憶功能障礙,但PCC組在傷后損傷程度較輕、恢復(fù)快;CCC組在傷后損傷程度較重、恢復(fù)較慢。2、腦震蕩大鼠中隔區(qū)、基底節(jié)、前額葉和中腦的ChAT活性表達(dá)有明顯變化;3、腦震蕩大鼠認(rèn)知障礙與相關(guān)腦區(qū)ChAT活性表達(dá)的變化有關(guān)聯(lián)。
[Abstract]:Objective: to observe the changes of cognitive impairment and acetylcholine transferase (acetylcholine transferase) in rats with cerebral concussion (Cerebral Concussion,CC) at different time and at different time after concussion, and to explore the cognition of concussion rats. The characteristics of memory disorder and its relationship with the changes of acetylcholine transferase (acetylcholine transferase), the neurobiological basis of cognitive memory disorder in concussion rats were understood, which provided theoretical basis for the clinical diagnosis and treatment of postconcussion syndrome. Methods: the rat model of cerebral concussion was established by using the mechanical striking device of metal monopodic closed brain injury. 70 SD rats were randomly divided into 8 groups: the rats with simple concussion group (Pure Cerebral Concussion,PCC) were divided into six groups (n = 10), complex concussion group (Complicated Cerebral Concussion,CCC) and normal control group (n = 10). Balance beam (Beam Balance test,BBT) and noise bar running test (Beam Walking test,BWT) were used to detect the changes of balance time (Balance Latency,BL) and incubation period (Step Through Latency,STL) of 1 day after injury. The changes of acetylcholine transferase (Cholin Acetyltransferase,CHAT) in the septum, basal ganglia, prefrontal lobe and midbrain of rats were detected by (Western Blot) on the 1st and 2nd day after PCC injury. Results: the behavioral results showed that there was no difference in balance time between PCC group and control group. However, in BWT, the incubation period was significantly prolonged only on 1 day and 2 days after injury. The balance time of CCC group was significantly different from that of control group on the 1st day after injury (p < 0. 05). In BWT, the incubation period was significantly prolonged on the 1st day, and gradually recovered from 48.12 鹵18.24 seconds to 27.10 鹵28.36 seconds. The results of immunoblotting showed that the expression of ChAT in basal ganglia began to rise on the second day after injury compared with the control group (p < 0. 05), and the expression of ChAT decreased on the 24th day after injury, but still did not return to normal. The ChAT in the septal area began to rise on the 1st day after injury, but the difference was not significant (p > 0. 05), but there was a significant difference up to the 4th day after injury (p < 0. 05). The ChAT of prefrontal lobe began to decrease on the 1st day after injury, but the difference was not significant (p > 0. 05), but the difference was significant up to the 8th day after injury (p < 0. 05). On the second day after ChAT injury, the difference was significant (p < 0. 05), and the difference was significant up to the 24th day after injury (p < 0. 05). Conclusion: 1. In the BBT and BWT experiments, the rats with cerebral concussion had different degrees of learning and memory impairment, but in the PCC group, the degree of injury was mild and the recovery was faster. In CCC group, the degree of injury was severe and the recovery was slower. 2. The expression of ChAT activity in septum, basal ganglia, prefrontal lobe and midbrain of rats with cerebral concussion showed significant changes. 3. Cognitive impairment was associated with the change of ChAT activity in related brain regions in concussion rats.
