新的STR遺傳標(biāo)記及熒光標(biāo)記復(fù)合擴(kuò)增的法醫(yī)學(xué)應(yīng)用研究

【摘要】:目的 本課題旨在尋找應(yīng)用于法醫(yī)的STR基因座并建立D20S601、D6S474、D6S2418熒光標(biāo)記復(fù)合擴(kuò)增體系,按照美國(guó)DNA分析方法技術(shù)工作組(The Technology Working Group on DNA Analysis Methods,TWGDAM)的指導(dǎo)方案進(jìn)行法醫(yī)學(xué)實(shí)用性研究。方法 對(duì)5個(gè)新的STR基因座進(jìn)行群體遺傳學(xué)研究,并對(duì)D20S601、D6S474、D6S2418基因座原始引物進(jìn)行了改進(jìn),利用傳統(tǒng)復(fù)合擴(kuò)增技術(shù)擴(kuò)增,ABI PRISM~(?)310基因分析儀對(duì)產(chǎn)物進(jìn)行檢測(cè);用混合樣本、不同的組織、不同的介質(zhì)、常見(jiàn)的動(dòng)物在不同的模板量、不同的溫度條件下對(duì)復(fù)合擴(kuò)增系統(tǒng)進(jìn)行測(cè)試,用實(shí)際案例進(jìn)行驗(yàn)證。結(jié)果 獲得5個(gè)STR基因座的基因頻率、個(gè)人識(shí)別率、非父排除率等群體遺傳學(xué)數(shù)據(jù);通過(guò)采用傳統(tǒng)的復(fù)合擴(kuò)增的方法,建立了D20S601、D6S474、D6S2418熒光標(biāo)記復(fù)合擴(kuò)增
[Abstract]:Objective the purpose of this study was to search for the STR loci used in forensic medicine and to establish a multiplex amplification system for D20S601, D6S474, D6S2418 fluorescent labeling. According to the (The Technology Working Group on DNA Analysis Methods, of the DNA analysis technique working group of the United States, TWGDAM) is used to study the practicability of forensic medicine. Methods five new STR loci were studied in population genetics. The primordial primers of D20S601D6S474nD6S2418 locus were improved. The products were amplified by traditional multiplex amplification technique and detected by ABI PRISM~ (?) 310 gene analyzer. The compound amplification system was tested with mixed samples, different tissues, different media, common animals under different template quantity and different temperature, and verified by actual cases. Results the population genetic data of 5 STR loci were obtained, such as gene frequency, personal identification rate, non-paternal exclusion rate, etc. By using the traditional multiplex amplification method, the fluorescent multiplex amplification of D20S601D6S474hD6S2418 was established.
