大鼠心肌和骨骼肌肌鈣蛋白I降解與死亡時間關(guān)系的研究

【摘要】: 目的 研究大鼠心肌肌鈣蛋白I(Cardiac troponin I, cTnI)和骨骼肌肌鈣蛋白I(Skeletal troponin I, sTnI)在體和離體兩種情況下的降解規(guī)律并進行比較,探討根據(jù)cTnI及sTnI降解情況推斷死亡時間的可行性,為死亡時間(Postmortem interval,PMI)推斷的研究提供新的思路和方法,為法醫(yī)實踐中PMI的推斷提供科學依據(jù)。 方法 將健康SD大鼠10只按在體和離體二種處置方式隨機平均分為2組,每組5只。將10只大鼠用機械性窒息法致死,取離體組大鼠心肌及骨骼肌組織置于保濕盒內(nèi)與在體組大鼠尸體一起放入人工氣候箱中,恒溫30℃,濕度恒定為40%保存。分別于取材后0、24、48、72、96和120h提取離體及在體組織中的蛋白質(zhì),應(yīng)用免疫印跡(Western blot)法結(jié)合圖像分析技術(shù)半定量檢測當時死亡及離體時間的大鼠cTnI、sTnI含量。用死后120h時cTnI或sTnI的含量占死亡即刻cTnI或sTnI的含量的百分數(shù)來表示cTnI及sTnI降解的相對速度。將所得數(shù)據(jù)進行統(tǒng)計學分析,尋找隨著PMI的延長各含量參數(shù)的變化規(guī)律,計算各參數(shù)與PMI的相關(guān)系數(shù),求出回歸方程。最后根據(jù)分析結(jié)果比較在體和離體cTnI或sTnI降解規(guī)律之間的異同。 結(jié)果 1、在體及離體情況下大鼠cTnI與sTnI的含量均隨死亡或離體時間延長而逐漸下降,且與死亡或離體時間的對數(shù)值有很強的線性關(guān)系。 2、在體情況下cTnI與sTnI的降解比離體情況慢。 3、在體和離體兩種情況下sTnI均比cTnI降解慢。 結(jié)論 1、cTnI和sTnI有望用于推斷五天以內(nèi)的死后經(jīng)過時間。 2、cTnI和sTnI在離體與在體條件下的降解情況并不完全一致,離體時的降解速度要比在體時快。 3、sTnI較cTnI更適用于推斷PMI。
