基于mRNA穩(wěn)定性推斷死亡時(shí)間的研究

【摘要】:目的探討大鼠死后不同時(shí)間腎、脾3-磷酸甘油醛脫氫酶(GAPDH)mRNA的表達(dá)對死亡時(shí)間(PMI)推斷的應(yīng)用價(jià)值。方法28只大鼠斷頸處死后置于20℃溫控系統(tǒng)內(nèi),分別于死后0,8,16,24,32,40,48h共7個(gè)時(shí)相點(diǎn)(n=4)解剖,利用熒光RT-PCR技術(shù)檢測大鼠腎、脾GAPDHmRNA相對表達(dá)量的變化。結(jié)果在大鼠死后40h腎、32h脾內(nèi)可檢測出GAPDHmRNA,且其表達(dá)產(chǎn)物均呈規(guī)律性下降趨勢(熒光強(qiáng)度逐漸變暗并最終消失)。結(jié)論檢測大鼠死后不同時(shí)間腎、脾GAPDHmRNA的表達(dá)對PMI推斷具有一定的應(yīng)用價(jià)值。
