熒光標(biāo)記RT-PCR技術(shù)在實(shí)驗(yàn)動(dòng)物死亡時(shí)間推斷中的應(yīng)用

【摘要】:目的探討熒光標(biāo)記逆轉(zhuǎn)錄-聚合酶聯(lián)反應(yīng)(RT-PCR)在死亡時(shí)間(PM I)推斷中的應(yīng)用。方法44只大鼠斷頸處死后置于20℃溫度控制系統(tǒng)內(nèi),利用熒光標(biāo)記RT-PCR技術(shù)檢測(cè)大鼠腦及肝組織GAPDH mRNA在死后即刻至8 d的相對(duì)表達(dá)量的變化。結(jié)果在大鼠死后即刻至7 d腦組織、2 d肝組織內(nèi)均可檢測(cè)到GAPDH mRNA,其擴(kuò)增產(chǎn)物的降解呈現(xiàn)出一定的規(guī)律性。結(jié)論熒光RT-PCR技術(shù)檢測(cè)大鼠死后腦及肝GAPDH mRNA,可為PM I推斷提供一種新的方法。
