熒光標記RT-PCR技術在實驗動物死亡時間推斷中的應用

【摘要】:目的探討熒光標記逆轉錄-聚合酶聯(lián)反應(RT-PCR)在死亡時間(PM I)推斷中的應用。方法44只大鼠斷頸處死后置于20℃溫度控制系統(tǒng)內(nèi),利用熒光標記RT-PCR技術檢測大鼠腦及肝組織GAPDH mRNA在死后即刻至8 d的相對表達量的變化。結果在大鼠死后即刻至7 d腦組織、2 d肝組織內(nèi)均可檢測到GAPDH mRNA,其擴增產(chǎn)物的降解呈現(xiàn)出一定的規(guī)律性。結論熒光RT-PCR技術檢測大鼠死后腦及肝GAPDH mRNA,可為PM I推斷提供一種新的方法。
