大鼠腦組織缺血GFAP、HSP70的表達(dá)與缺血時(shí)間推斷的研究

【摘要】:目的探討大鼠腦組織缺血后不同時(shí)間膠質(zhì)原纖維酸性蛋白(Glial fibrillary acidic protein,GFAP)和熱休克蛋白70(Heat shock protein 70,HSP70)的表達(dá),旨為法醫(yī)實(shí)踐中推斷腦組織缺血時(shí)間提供科學(xué)的參考依據(jù)。 方法建立Wistar 大鼠腦組織缺血模型,應(yīng)用免疫組化S-P 法與顯微圖像分析技術(shù)檢測(cè)腦組織缺血后10min、30min、1h、3h、6h、12h、24h、48h 和正常組GFAP、HSP70 的表達(dá),觀察是否有統(tǒng)計(jì)學(xué)意義。 結(jié)果(1)大鼠海馬區(qū)神經(jīng)細(xì)胞缺血3h 可見水腫、壞死,錐體細(xì)胞受損;缺血48h 見大量錐體細(xì)胞壞死,核膜崩解、核仁消失而僅留細(xì)胞碎屑。(2)腦組織缺血3h,GFAP 在星形膠質(zhì)細(xì)胞的陽(yáng)性表達(dá)與正常組比較無差異; 6h 與3h 比較有差異(p 0.05);12-48h 各組間比較有顯著性差異(p 0.01)。(3)HSP70 的表達(dá)隨著缺血時(shí)間的延長(zhǎng)而逐漸增高,HSP70 陽(yáng)性細(xì)胞于腦缺血后10min 后開始出現(xiàn),隨著缺血時(shí)間的延長(zhǎng)開始逐漸增多,缺血6h 內(nèi)各組間比較無差異;6-12h 各組間比較有差異(p 0.05);24-48h各組間比較有顯著性差異(p 0.01)。 結(jié)論大鼠腦組織缺血GFAP、HSP70 陽(yáng)性表達(dá)呈現(xiàn)一定規(guī)律,與缺血時(shí)間明顯相關(guān),可作為推斷腦缺血時(shí)間的參考指標(biāo)之一,在法醫(yī)檢案中具有應(yīng)用價(jià)值。
[Abstract]:Objective to investigate the expression of Glial fibrillary acidic protein (GFAP) and heat shock protein 70 (Heat shock protein 70, HSP70) at different time after cerebral ischemia in rats, and to provide a scientific reference for the inference of cerebral ischemic time in forensic practice.
Methods the cerebral ischemia model of Wistar rats was established, and the immunohistochemical S-P method and microscopic image analysis technique were used to detect 10min, 30min, 1H, 3h, 6h, 12h, 24h, 48h and GFAP, HSP70, and whether there was statistical significance.
Results (1) edema, necrosis, and pyramidal cells were damaged in the ischemic 3H in the hippocampus of the rats, and the ischemic 48h saw a large number of pyramidal cells necrosis, the nuclear membrane disintegrating and the nucleolus disappeared and only the cell debris left. (2) the cerebral ischemia 3h, the positive expression of GFAP in astrocytes was not different from that in the normal group; 6h and 3H were different (P 0.05); 12-48h There were significant differences in each group (P 0.01). (3) the expression of HSP70 increased gradually with the prolongation of ischemic time. The HSP70 positive cells began to appear after 10min after cerebral ischemia, and gradually increased with the prolongation of ischemic time. There was no difference between each group in the ischemic 6h (P 0.05), and the ratio of 24-48h to each group was compared. There was a significant difference (P 0.01).
Conclusion the positive expression of GFAP and HSP70 in the cerebral ischemia of rats is related to the ischemic time. It can be used as one of the reference indexes to infer the time of cerebral ischemia, and it is valuable in forensic examination.
