單細胞檢驗技術用于混合上皮細胞的分離和檢驗

【摘要】:目的建立單細胞顯微捕獲聯(lián)合低體積擴增技術,用于混合上皮細胞檢材分離檢驗。方法取5名男性口腔上皮細胞拭子浸泡液30μL,分別滴加到5份含同一女性皮膚表皮細胞拭子上,制成5份混合上皮細胞樣本為實驗組,同時制備同樣的5份樣本為對照組。實驗組樣本采用顯微捕獲單個口腔上皮細胞,并使用低體積擴增技術進行擴增;對照組用M48純化試劑盒提取DNA,Identifiler試劑盒復合擴增,擴增體系為10μL。所有產物均用ABI 3130遺傳分析儀進行STR分型。結果 5份實驗組樣本均得到男性STR分型結果,5份對照組樣本則均僅得到混合分型結果。將該方法應用于1例強奸殺人案例檢驗,取得了滿意效果。結論單細胞顯微捕獲聯(lián)合低體積擴增技術可用于混合上皮細胞樣本的分離檢驗。
[Abstract]:Objective to establish a technique of single cell microcapture combined with low volume amplification for the separation and detection of mixed epithelial cell samples. Methods the oral epithelial cell swabs of 5 male patients were treated with 30 渭 L oral epithelial cell swabs. Five samples of mixed epithelial cells were prepared as experimental group and the same 5 samples were prepared as control group. In the experimental group, a single oral epithelial cell was captured microscopically and amplified by low volume amplification, while in the control group, the DNA identifiler was extracted by M48 purification kit, and the amplification system was 10 渭 L. All the products were classified by ABI 3130 genetic analyzer. Results the results of STR typing were obtained from 5 experimental group samples and only mixed typing results were obtained from 5 control group samples. The method has been applied to a case of rape homicide, and satisfactory results have been obtained. Conclusion single cell microcapture combined with low volume amplification can be used to separate and test mixed epithelial cell samples.
