十個(gè)新的Y染色體STR的特性及復(fù)合擴(kuò)增研究

【摘要】:目的 本課題旨在提高Y染色體的鑒別能力,為法醫(yī)Y染色體分型提供新的遺傳標(biāo)記。對(duì)十個(gè)新的Y染色體STR基因座進(jìn)行序列研究和單倍型分析,為等位基因的命名奠定基礎(chǔ),為法醫(yī)學(xué)應(yīng)用提供概率計(jì)算依據(jù)。選擇其中四個(gè)基因座,依托毛細(xì)管電泳激光熒光檢測(cè)平臺(tái),應(yīng)用熒光標(biāo)記復(fù)合擴(kuò)增技術(shù),建立一套新的Y染色體STR基因座復(fù)合擴(kuò)增體系,并按照DNA分析方法技術(shù)工作組(The Technology Working Group on DNA Analysis Methods, TWGDAM)的指導(dǎo)方案進(jìn)行法醫(yī)學(xué)應(yīng)用研究,以評(píng)估其在法醫(yī)鑒定中的可用性。方法 選擇十個(gè)新Y-STR基因座,PCR擴(kuò)增四川成都漢族103個(gè)無關(guān)男性DNA樣本,PCR擴(kuò)增產(chǎn)物使用聚丙烯酰胺凝膠電泳分離,銀染法顯色,以獲得其群體遺傳數(shù)據(jù)。選取其中四個(gè)Y-STR基因座,用自行設(shè)計(jì)的熒光標(biāo)記引物復(fù)合擴(kuò)增,毛細(xì)管電泳熒光自動(dòng)檢測(cè)系統(tǒng)對(duì)復(fù)合擴(kuò)增產(chǎn)物進(jìn)行檢測(cè)和分型;用混合樣本、不同的組織樣本、不同介質(zhì)上的DNA樣本和常見動(dòng)物的DNA作為模板,在不同的條件下對(duì)復(fù)合擴(kuò)增系統(tǒng)進(jìn)行測(cè)試和優(yōu)化,用實(shí)際案例進(jìn)行驗(yàn)證。結(jié)果 在我們研究的103個(gè)成都漢族男性樣本中,十個(gè)Y染色體STR基因座各觀察到3~8個(gè)等位基因,等位
[Abstract]:Objective to improve the differentiation ability of Y chromosome and to provide a new genetic marker for forensic Y chromosome typing. The sequence study and haplotype analysis of ten new Y chromosome STR loci were carried out, which laid the foundation for allele naming and provided the basis of probability calculation for forensic application. Four of these loci were selected, and a new system of Y chromosome STR locus multiplex amplification was established based on capillary electrophoresis laser fluorescence detection platform and fluorescence labeling multiplex amplification technique. Forensic applied research was carried out under the guidance of the Technology working Group on DNA Analysis methods (TWGDAM) to assess its availability in forensic identification. Methods Ten new Y-STR loci were selected for PCR amplification of 103 unrelated male DNA samples from Sichuan Han nationality in Chengdu. Polyacrylamide gel electrophoresis was used to isolate the PCR products. Four Y-STR loci were selected for multiplex amplification with self-designed fluorescent labeled primers, and the multiplex amplification products were detected and typed by capillary electrophoresis fluorescence automatic detection system. DNA samples from different media and DNA from common animals were used as templates to test and optimize the multiplex amplification system under different conditions. Results among the 103 male samples of Chengdu Han nationality, 38 alleles and alleles were observed in 10 STR loci of Y chromosome.
