【摘要】：目的研究一次性使用牙刷上脫落細(xì)胞的DNA提取和STR分型。方法對一次性使用牙刷的采集方法、采集部位、DNA提取方法、存放時間對STR分型的影響進(jìn)行比對研究。結(jié)果割取法可獲得較高濃度的DNA,30例中檢出9個以上基因座達(dá)27例,與擦拭法存在統(tǒng)計(jì)學(xué)差異(P0.05)。Chelex-100法、DNA IQTM法檢出9個以上基因座分別為26例、24例,STR分型結(jié)果無統(tǒng)計(jì)學(xué)意義(P0.05)。提取牙刷的前、后部三束刷毛檢出9個以上基因座分別達(dá)27例、28例,STR分型結(jié)果無統(tǒng)計(jì)學(xué)意義(P0.05);放置1天、1周、1個月、3個月、6個月的時間后檢出9個以上基因座分別為28例、27例、22例、12例、7例。結(jié)論割取法提取一次性牙刷上的脫落細(xì)胞進(jìn)行STR分型效果良好;放置時間越長的牙刷,檢出率越低。
[Abstract]:Objective to study the DNA extraction and STR typing of exfoliated cells from disposable toothbrush. The effect of storage time on STR typing was compared. Results in the 30 cases with high concentration of DNAs, more than 9 loci were detected in 27 cases. There was a statistical difference between P0.05G. Chelex-100 DNA IQTM method and wiping method. More than 9 loci were detected in 26 cases and 24 cases, respectively. The results of STR typing were not statistically significant (P 0.05). Before and after the extraction of toothbrush, more than 9 loci were detected in 27 cases and 28 cases respectively. The results of STR typing were not statistically significant (P 0.05). After one day, one week, one month, three months and six months, more than 9 loci were detected in 28 cases, 27 cases, 22 cases and 12 cases, respectively. Conclusion extraction of exfoliated cells from disposable toothbrush is effective for STR typing. The longer the toothbrush is placed, the lower the detection rate is.
