中國大鯢ERα、P450arom基因的克隆及不同發(fā)育期4種性別決定相關(guān)基因的表達分析

中國大鯢(Andrias davidianus)是我國特有的有尾兩棲動物,也是全球現(xiàn)存兩棲類中體型最大的古老物種,屬于國家Ⅱ級重點保護野生動物,具有重要的經(jīng)濟、科研和生態(tài)價值。然而,近年來由于棲息地的破壞、過度捕殺以及傳染病等因素,導(dǎo)致中國大鯢種群數(shù)量急劇下降。為了更好地保護這一古老物種,我國科研人員開始了中國大鯢的人工馴養(yǎng)繁殖研究,先后形成了中國大鯢全人工繁育(Captive Breeding)、原生態(tài)繁育(Natural Eco-breeding)、仿生態(tài)繁育(Imitated Natural Eco-breeding)等3種人工繁殖技術(shù)。隨著人工繁育技術(shù)的突破,中國大鯢人工種群數(shù)量劇增。然而,在人工較高水溫飼養(yǎng)條件下,幼鯢絕大部分發(fā)育成為雄性,造成中國大鯢人工種群性別比例嚴(yán)重失調(diào)。為了解決此問題,在幼鯢性腺分化的敏感期,使用一定濃度的外源雌二醇斷續(xù)暴露40 d,又使得幼鯢已連續(xù)4年全部分化為雌性。以上現(xiàn)象表明,中國大鯢性腺分化受到環(huán)境水溫與外源雌二醇的直接影響。本文在中國大鯢首次克隆了雌激素受體(estrogen receptor alpha, ERa)、細胞色素P450芳香化酶...
