中華鱉輸卵管Telocyte的特性研究

輸卵管是脊椎動物重要的生殖器官,參與和調(diào)控卵子的運輸、成熟、儲存、受精與早期胚胎發(fā)育,但各種動物輸卵管的組成與結(jié)構(gòu)存在差異。中華鱉是我國養(yǎng)殖面積最大的爬行動物,不僅具有重要的經(jīng)濟價值,而且具有特殊的精子儲存現(xiàn)象。中華鱉輸卵管由漏斗部、蛋白分泌部、峽部、子宮部和陰道部5段組成,其中峽部、子宮部和陰道部前段是發(fā)生精子儲存的部位。新近在哺乳動物器官中發(fā)現(xiàn)了一種特異的細胞—Telocyte(暫時翻譯為遠細胞),參與體內(nèi)許多器官(尤其是管狀器官)的功能活動與調(diào)節(jié)。但Telocyte在非哺乳動物的分布與作用未見資料報道。本文利用形態(tài)學方法和細胞生物學技術(shù),觀測鑒定Telocyte在中華鱉輸卵管精子儲存部位(包括峽部、子宮部和陰道部)的分布,分析其超微結(jié)構(gòu)特點,闡明Telocyte及其突起與周圍環(huán)境和細胞的形態(tài)聯(lián)系。進一步在分子水平上探究Telocyte標記物-CD34 mRNA在輸卵管的表達。首次在哺乳類之外的動物體內(nèi)鑒定發(fā)現(xiàn)telocyte的分布及其特性,研究結(jié)果為闡明中華鱉精子儲存機理提供細胞分子基礎(chǔ)。實驗Ⅰ中華鱉輸卵管陰道部telocytes的超微結(jié)構(gòu)與免疫組化特征Telocytes是2005年新發(fā)現(xiàn)的一種間質(zhì)細胞,由Popescu和他的同事命名,目前已經(jīng)在哺乳動物不同類型的組織和器官中被確認存在,但陰道部中未見資料報道(www.telocytes.com)。本研究利用透射電鏡和免疫組化探查中華鱉輸卵管陰道部telocytes的分布、超微結(jié)構(gòu)以及它們與周圍細胞的形態(tài)關(guān)系。結(jié)果顯示,telocytes主要位于單層柱狀上皮下方結(jié)締組織中,靠近分泌腺細胞處可見Tcs及其突起,有些Tcs及其突起與肥大細胞和膠原纖維密切相關(guān)。Tcs及其突起呈CD34免疫反應(yīng)陽性。在細胞水平上顯示Tcs在中華鱉輸卵管陰道部的定位,它們的細胞標記與哺乳類具有同源性。研究結(jié)果為理解陰道功能及防治陰道疾病提供了新視角。實驗Ⅱ中華鱉輸卵管子宮部telocytes的超微特征Telocytes(Tcs)與體內(nèi)許多機能調(diào)節(jié)和某些疾病發(fā)生有關(guān)。Tcs具有極其細長的突起(telopodes,Tps),可伸達遠端,有人將其翻譯為遠(端)細胞。Tps由細段(podomers)和膨大部(podoms)交替排列,呈串珠樣結(jié)構(gòu)。Tcs的研究報道全部為人和哺乳動物的資料,已經(jīng)在心、肺、腎、消化道等器官組織中被鑒定發(fā)現(xiàn)。應(yīng)用透射電鏡對中華鱉子宮部Tcs的超微結(jié)構(gòu)特性進行了觀察鑒定,結(jié)果顯示中華鱉輸卵管子宮部具有Tcs分布,而且主要位于子宮固有膜結(jié)締組織中,胞體結(jié)構(gòu)完整,突起細長,與神經(jīng)末梢、毛細血管、膠原纖維束以及子宮腺密切相關(guān)。這種結(jié)構(gòu)關(guān)系表明,Tcs及其突起具有參與中華鱉子宮收縮及其腺體分泌的形態(tài)學特征。實驗Ⅲ中華鱉輸卵管峽部telocytes的特征Telocytes擁有一個很小的細胞體,很細長的telopodes(Tps),且podomers與podoms交替排布。Tps釋放3種胞外小泡:外來體,核外顆粒體及多泡小體,這些都參與旁分泌信號傳導。本研究通過透射電鏡及免疫組化研究中華鱉輸卵管峽部內(nèi)telocytes的超微形態(tài)特征及其結(jié)構(gòu)功能。峽部具有不同形狀的telocytes,這些形態(tài)取決于突起的數(shù)量。分布位置與輸卵管子宮部相似,一般位于黏膜固有層。重要的發(fā)現(xiàn)是,telocytes參與細胞外囊泡的分泌和轉(zhuǎn)運。該研究第一次揭示了中華鱉輸卵管峽部內(nèi)TCs的存在及其分布,并在超微水平上顯示Tcs具有分泌和轉(zhuǎn)運細胞外囊泡的功能。實驗ⅣCD34 mRNA在中華鱉輸卵管的表達造血祖細胞CD34 Telocytes的一種特異標記物。本文應(yīng)用qRT-PCR技術(shù)分析了CD34-mRNA在中華鱉輸卵管的表達動態(tài)。CD34基因表達于輸卵管蛋白分泌部、峽部、子宮部和陰道部4段,與之對應(yīng),透射電鏡下大量的Telocytes細胞分布于中華鱉輸卵管壁內(nèi)。CD34 mRNA的表達水平具有區(qū)域性差異,子宮部最高,陰道部次之,峽部中等,而蛋白分泌部表達最低。這種表達區(qū)域性差異可能與輸卵管節(jié)律性蠕動的調(diào)控有關(guān)。
