【摘要】：類(lèi)黃酮是一類(lèi)結(jié)構(gòu)多樣(超過(guò)10,000種),具有廣泛生物活性的植物次生代謝物。黃酮是類(lèi)黃酮的亞家族之一,功能非常重要但是研究不多。代謝物的糖基化修飾對(duì)植物細(xì)胞的新陳代謝至關(guān)重要,而黃酮的糖基化修飾由UDP-葡萄糖糖基轉(zhuǎn)移酶負(fù)責(zé)完成。代謝物的不同的修飾能夠極大地增加自然界代謝物的多樣性,但是這種修飾對(duì)代謝物的生物學(xué)功能有何影響還有待研究。本研究中,我們證明了水稻中氧黃酮的自然變異主要由黃酮5-O-糖基轉(zhuǎn)移酶(OsUGT707A2)和黃酮7-O-糖基轉(zhuǎn)移酶(OsUGT706D1)控制。借助體內(nèi)、體外分析和轉(zhuǎn)基因植株的紫外處理實(shí)驗(yàn),我們進(jìn)一步證明了這兩個(gè)基因能夠提高水稻對(duì)UV-B的耐受性。除此之外,我們還克隆水稻中4個(gè)控制碳黃酮糖基化的糖基轉(zhuǎn)移酶(Os06g18010,Os06g18140,Os06g18670和Os06g18790),并在大腸桿菌中對(duì)上述4個(gè)蛋白進(jìn)行了原核表達(dá),純化。我們還通過(guò)體外酶活測(cè)定實(shí)驗(yàn)以及轉(zhuǎn)基因水稻代謝物分析實(shí)驗(yàn)進(jìn)一步確定這些基因在水稻體內(nèi)的實(shí)際功能。Os06g18010和Os06g18140不僅可以利用根皮素作為底物產(chǎn)生對(duì)應(yīng)的碳糖基化黃酮,還可以利用芹菜素作為底物產(chǎn)生氧糖基化黃酮,但是Os06g18670和Os06g18790不能利用根皮素作為底物。對(duì)Os06g18670和Os06g18790的超表達(dá)植株進(jìn)行UV處理,我們發(fā)現(xiàn),相比野生型水稻,這些超表達(dá)水稻植株具有更強(qiáng)的紫外耐受性。以上結(jié)果不僅有利于解析黃酮合成的生化途徑和遺傳調(diào)控,同時(shí)也說(shuō)明將優(yōu)良等位基因應(yīng)用到育種中這個(gè)思路是改善農(nóng)作物產(chǎn)量的重要一環(huán)。
