反相高效液相色譜法同時測定新生兒尿液中4種苯丙酮尿癥特征酸的含量

【摘要】:提出了反相高效液相色譜同時測定新生兒尿液中4-羥基苯乙酸、2-羥基苯乙酸、苯乙酸和苯丙酮酸等4種苯丙酮尿癥特征酸的方法。取尿液樣品5.00mL,乙腈5mL,混合振蕩1min,離心10min,取上清液進行液相色譜分離。用Waters Symmetry C18色譜柱作為固定相,用不同比例的乙腈和0.15%(體積分數(shù))磷酸溶液進行梯度洗脫,檢測波長為210,289nm。在上述條件下,4種特征酸分離完全,并且不受尿液中內(nèi)源性物質(zhì)的干擾。4種特征酸的質(zhì)量濃度在一定范圍內(nèi)呈線性,檢出限(3S/N)分別為0.43,0.22,0.62,0.42mg·L-1。按標準加入法在3個濃度水平上進行加標回收試驗,測得回收率在89.1%~108%之間,測定值的相對標準偏差(n=6)小于4%。
[Abstract]:A reverse-phase high performance liquid chromatography for the determination of 4-hydroxyphenylacetic acid,2-hydroxyphenylacetic acid, phenylacetic acid and phenylacetone acid in the urine of the newborn was presented. 5.00 mL of urine sample,5 mL of acetonitrile,1 min of mixing, and 10 min for centrifugation, and the supernatant was separated by liquid chromatography. The gradient elution was performed with a Waters Symmetry C18 column as a stationary phase with a different ratio of acetonitrile and 0.15% (volume fraction) phosphoric acid solution, with a detection wavelength of 210,289 nm. Under the above conditions,4 kinds of characteristic acid were separated completely and not interfered by endogenous substances in urine. The mass concentration of 4 kinds of characteristic acids was linear in a certain range, and the detection limit (3S/ N) was 0.43, 0.22, 0.62 and 0.42 mg 路 L-1, respectively. The recoveries were found to be between 89.1% and 108%, and the relative standard deviation (n = 6) of the measured values was less than 4%.
