【摘要】：目的對(duì)臨床分離于皮脂腺囊腫標(biāo)本中的菌株進(jìn)行表型和基因型鑒定,描述該菌株的生物學(xué)特性及致病性,為臨床診斷提供準(zhǔn)確地病原學(xué)依據(jù)。方法對(duì)分離菌株進(jìn)行革蘭氏染色等,并使用VITEK 2Compact全自動(dòng)微生物分析儀進(jìn)行鑒定。采用K-B法進(jìn)行藥敏試驗(yàn)。提取分離菌株DNA,采用通用引物對(duì)16S rRNA基因進(jìn)行PCR擴(kuò)增并測(cè)序,將測(cè)序結(jié)果與GenBank中收錄的16S rRNA基因序列進(jìn)行BLAST同源性對(duì)比。結(jié)果分離菌株經(jīng)VITEK 2Compact鑒定為玫瑰色庫(kù)克菌,后經(jīng)16S rRNA序列測(cè)定方法鑒定,分離菌株為產(chǎn)丙酮酸棒狀桿菌。藥敏試驗(yàn)顯示該菌株對(duì)四環(huán)素、萬(wàn)古霉素、利福平敏感,對(duì)青霉素、紅霉素、克林霉素、環(huán)丙沙星、左氧氟沙星、復(fù)方新諾明、頭孢吡肟、亞胺培南、慶大霉素等均耐藥。結(jié)論對(duì)于表現(xiàn)型不易鑒定或鑒定不準(zhǔn)確的細(xì)菌,采用16SrRNA序列測(cè)定的方法進(jìn)行鑒定是最準(zhǔn)確的。產(chǎn)丙酮酸棒狀桿菌是引起患者疾病的致病菌。在完善產(chǎn)丙酮酸棒狀桿菌生化反應(yīng)信息的同時(shí),探索出有效可行的生物學(xué)鑒定方法。
[Abstract]:Aim to identify the phenotypes and genotypes of the strains isolated from sebaceous gland cyst specimens and to describe the biological characteristics and pathogenicity of the strains in order to provide accurate etiological basis for clinical diagnosis. Methods the isolated strains were stained by Gram and identified by VITEK 2Compact automatic microbiological analyzer. K B method was used for drug sensitivity test. The 16s rRNA gene was amplified and sequenced by PCR with universal primers from the isolated strain DNA,. The BLAST homology of the 16s rRNA gene was compared with that of the 16s rRNA gene included in GenBank. Results the isolated strain was identified as Corynebacterium roseus by VITEK 2Compact and then identified by 16s rRNA sequencing. The isolated strain was Corynebacterium pyruvate-producing. The susceptibility test showed that the strain was sensitive to tetracycline, vancomycin, rifampicin, and resistant to penicillin, erythromycin, clindamycin, ciprofloxacin, levofloxacin, compound sinomine, cefepime, imipenem and gentamicin. Conclusion 16SrRNA sequencing is the most accurate method for identification of bacteria which are not easy to be identified or not accurately identified. Corynebacterium pyruvate-producing bacteria is the cause of disease in patients. In order to improve the biochemical reaction information of Corynebacterium pyruvate-producing bacteria, an effective and feasible biological identification method was explored.
