具有光動力治療效果的超分子聚合物比率式納米氧氣傳感器

【摘要】:低氧是許多疾病產(chǎn)生的信號,例如癌癥、心血管疾病、中風(fēng)、阿茲海默病和帕金森病等,所以及時檢測人體細(xì)胞和組織中氧氣對癌癥的診斷至關(guān)重要;诹坠忖绲谋嚷适焦鈱W(xué)氧氣傳感器靈敏度高、選擇性好、可逆性好以及檢測時無需消耗氧氣受到廣大科研工作者的關(guān)注。光動力療法(PDT)技術(shù)相比化學(xué)療法具有副作用小、抗藥性小和低毒性等優(yōu)點,對癌癥的治療極具潛力。因此,發(fā)展具有光動力治療效果的多功能氧氣傳感器具有重要的意義。超分子聚合物由小分子單體通過可逆的非共價鍵作用相連接共聚而成,其組份和功能可以通過單體結(jié)構(gòu)的改變來調(diào)節(jié),為構(gòu)筑比率式光學(xué)氧氣傳感器提供了基礎(chǔ)。本論文開展了以下三部分的研究:1、合成了具有雙四重氫鍵的磷光化合物Por(Pt)-bisUPy和熒光化合物DPA-bisUPy,它們的結(jié)構(gòu)通過核磁共振氫譜、核磁共振碳譜及高分辨質(zhì)譜進行了表征,這兩種化合物的光物理性質(zhì)能較好滿足構(gòu)筑比率式氧氣傳感器的要求。2、通過核磁氫譜和粘度測試證明了化合物DPA-bisUPy和Por(Pt)-bisUPy在氯仿中可以形成超分子聚合物鏈;通過微乳液法制備了超分子聚合物納米球NP1、NP2、NP3,通過SEM和TEM表征,表明這三種超分子聚合物納米球具有規(guī)整的形貌與尺寸,粒徑大約為60 nm左右;能量給體DPA-bisUPy與能量受體Por(Pt)-bisUPy之間存在高效的能量傳遞,進而顯著提高氧氣傳感器的磷光量子效率;氧氣猝滅研究結(jié)果表明該傳感器對氧氣具有高的靈敏度,達(dá)到了生物學(xué)測試的要求,它不僅是比率式還是比色式光學(xué)氧氣傳感器。3、超分子聚合物比率式納米氧氣傳感器具有良好的光穩(wěn)定性以及較好的生物相容性,可用于光動力治療和細(xì)胞成像。
[Abstract]:Hypoxia is the signal of many diseases, such as cancer, cardiovascular disease, stroke, Alzheimer's disease and Parkinson's disease, so the timely detection of oxygen in human cells and tissues is essential for the diagnosis of cancer. The ratio optical oxygen sensor based on phosphorescence quenching is of high sensitivity, good selectivity, good reversibility and no oxygen consumption in detection. Compared with chemotherapy, photodynamic therapy (PDT) has the advantages of less side effect, less drug resistance and less toxicity, so it has great potential in the treatment of cancer. Therefore, it is of great significance to develop a multifunctional oxygen sensor with photodynamic effect. Supramolecular polymers are composed of small molecular monomers linked and copolymerized by reversible noncovalent bonding. Their composition and function can be adjusted by the change of monomer structure, which provides the basis for the construction of proportional optical oxygen sensors. In this thesis, we have studied the following three parts: 1, and synthesized phosphorescence compound Por (Pt) bisUPy with double quadruple hydrogen bond and fluorescent compound DPA-bisUPy,. Their structures were characterized by NMR, NMR carbon and high resolution mass spectrometry. The photophysical properties of these two compounds can meet the requirements of constructing a ratio oxygen sensor. The results of NMR and viscosity measurements show that the compounds DPA-bisUPy and Por (Pt) bisUPy can form supramolecular polymer chains in chloroform. The supramolecular polymer nanospheres (NP1,NP2,NP3,) were prepared by microemulsion method and characterized by SEM and TEM. The results show that the three supramolecular polymer nanospheres have regular morphology and size, and the particle size is about 60 nm. There is a high energy transfer between the energy donor DPA-bisUPy and the energy receptor Por (Pt) bisUPy, which can significantly improve the phosphorescence quantum efficiency of the oxygen sensor, and the oxygen quenching results show that the sensor has a high sensitivity to oxygen. It is not only a ratio or colorimetric optical oxygen sensor .3. the supramolecular polymer ratio nano-oxygen sensor has good photostability and good biocompatibility. It can be used in photodynamic therapy and cell imaging.
