【摘要】：目的:通過VITEK MS微生物檢測系統(tǒng)對養(yǎng)殖場及市售活雞來源的空腸彎曲菌和單核細胞性李斯特菌進行激光解析飛行質(zhì)譜鑒定,建立兩種致病菌的質(zhì)譜鑒定方法。方法:從樣品中分離獲得疑似的空腸彎曲菌和單核細胞性李斯特菌,經(jīng)不同配比的基質(zhì)液處理后進行質(zhì)譜鑒定。通過分析檢出菌特征峰及相對豐度差異來評價不同配比基質(zhì)液對兩種致病菌檢測的影響。結(jié)果:空腸彎曲菌和李斯特菌對基質(zhì)液配比要求具有明顯差異,前者不需要經(jīng)過甲酸裂解,而對三氟乙酸含量敏感,后者鑒定結(jié)果受到甲酸裂解制約。基質(zhì)液含水量控制在體積分數(shù)30%可以有效避免特征峰缺失并提高檢出率。結(jié)論:控制基質(zhì)液的含水量、基質(zhì)濃度和三氟乙酸含量可以增加空腸彎曲菌和李斯特菌的特征峰數(shù)量,增強相對豐度,提高致病菌檢出率。基質(zhì)液配比優(yōu)化研究有助于增強食源性致病菌快速鑒定能力,有助于提高疾病預(yù)防和食源性風(fēng)險監(jiān)測的時效性。
[Abstract]:Objective: to identify Campylobacter jejuni and Listeria monocytogenes from poultry farms and live chickens by VITEK MS microbiological detection system, and to establish two methods for identification of pathogenic bacteria. Methods: the suspected Campylobacter jejuni and Listeria monocytogenes were isolated from the samples. By analyzing the characteristic peaks and the relative abundance differences, the effects of different proportion of matrix solution on the detection of two pathogenic bacteria were evaluated. Results: there was significant difference between Campylobacter jejuni and Listeria in the proportion of matrix solution. The former was sensitive to the content of trifluoroacetic acid without formic acid cracking, and the latter was restricted by formic acid cracking. Controlling the water content of matrix solution at 30% of volume fraction can effectively avoid the absence of characteristic peak and increase the detection rate. Conclusion: controlling the water content, the concentration of matrix and the content of trifluoroacetic acid can increase the number of characteristic peaks of Campylobacter jejuni and Listeria jejuni, enhance the relative abundance and increase the detection rate of pathogenic bacteria. The optimization of matrix ratio is helpful to enhance the ability of rapid identification of foodborne pathogens and to improve the timeliness of disease prevention and foodborne risk monitoring.
