【摘要】:目的建立電感耦合等離子體質(zhì)譜(inductively coupled plasma mass spectrometry,ICP-MS)測定人臍血清中22種元素含量的方法。方法吸取血清樣本以2%硝酸15倍稀釋,5000 r/min離心5 min,取上清液進樣分析。用Ge作為內(nèi)標元素校正八極桿碰撞模式定量元素(Fe、As、Se、I),用Sc、Ge、In、Bi作為標準模式定量元素的內(nèi)標。ICP-MS射頻功率1550 W,等離子氣體流量15.00 L/min,進樣量0.30 mL/min,駐留時間90 ms;八極桿反應(yīng)氣體為高純He氣,氣體流速1.0 L/min,射頻功率150 V,能量歧視5.0 V。結(jié)果內(nèi)標標準曲線法定量,各元素在其線性范圍內(nèi)線性良好(r0.9993),檢出限為0.00158~1.21μg/L。質(zhì)控血清測得值在標準值范圍內(nèi)。方法用于實際血清測定,加標回收率在86.4%~116%,測定精密度在0.164%~18.79%(n=6)。結(jié)論 ICP-MS法快速、靈敏、準確,操作簡單,能有效地檢測臍血清中各元素含量。
[Abstract]:Objective to establish an inductively coupled plasma mass spectrometry (ICP-MS) method for the determination of 22 elements in human umbilical serum. Methods the serum samples were centrifuged with 2% nitric acid 15 times dilution of 5 000 r/min for 5 min and the supernatant was analyzed. Using GE as the internal standard element to correct the quantitative element of the ocpole collision mode, using ScGe Ge Inn Bi as the internal standard of the quantitative element of the standard mode, ICP-MS RF power 1550 W, plasma gas flow rate 15.00 L / min, injection amount 0.30 mL / min, residence time 90 Ms, and the reaction gas of the eight pole is high purity he gas, Gas flow rate 1.0 L / min, RF power 150 V, energy discrimination 5.0 V. Results the standard curve of internal standard was used to determine the quantity. The linear range of each element was good. The detection limit was 0.00158 ~ 1.21 渭 g 路L ~ (-1) 路L ~ (-1) 路L ~ (-1) 路L ~ (-1) ~ (-1) ~ (-1). The value of quality control serum is in the range of standard value. The method has been applied to the determination of actual serum. The recovery rate of the added standard is 86.4 and the precision of the determination is 0.164 and 18.79. Conclusion ICP-MS method is rapid, sensitive, accurate and easy to operate, and can be used for the determination of all elements in umbilical serum.
