不同腫瘤細(xì)胞內(nèi)源性DNA雙鏈斷裂水平與放射敏感性參數(shù)SF2的相關(guān)性

【摘要】:目的探討不同腫瘤細(xì)胞內(nèi)源性DNA雙鏈斷裂(DSBs)水平與放射敏感性參數(shù)SF2是否存在相關(guān)性。方法選取肺癌細(xì)胞系H1299、A549,乳腺癌細(xì)胞系MCF-7、MDA-MB-435s為研究對(duì)象。利用2 Gy射線照射后克隆形成實(shí)驗(yàn)檢測(cè)不同細(xì)胞的放射敏感性指標(biāo)SF2;通過(guò)實(shí)時(shí)熒光定量PCR方法測(cè)定不同細(xì)胞相對(duì)端粒長(zhǎng)度變化;通過(guò)免疫熒光標(biāo)記法標(biāo)記不同腫瘤細(xì)胞γ-H2AX蛋白,檢測(cè)不同細(xì)胞內(nèi)源性DSBs損傷水平。通過(guò)統(tǒng)計(jì)學(xué)分析評(píng)價(jià)腫瘤細(xì)胞內(nèi)源性DSBs與放射敏感性的關(guān)系。結(jié)果免疫熒光法標(biāo)記γ-H2AX蛋白顯示4種腫瘤細(xì)胞內(nèi)源性DSBs水平腫瘤細(xì)胞除H1299與A549之間差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)外,其他任意兩組之間差異均有統(tǒng)計(jì)學(xué)意義(P0.05);4種腫瘤細(xì)胞的內(nèi)源性DSBs水平與SF2、端粒長(zhǎng)度的相關(guān)系數(shù)R分別為-0.95和-0.97,有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論腫瘤細(xì)胞內(nèi)源性DSBs可以作為一種預(yù)測(cè)腫瘤細(xì)胞放射敏感性的標(biāo)志,它們之間的相關(guān)性可能與端粒部分參與了內(nèi)源性DSBs產(chǎn)生和修復(fù)有關(guān)。
[Abstract]:Objective to investigate the correlation between the levels of endogenous DNA double strand breaks (DSBs) and radiosensitivity parameters (SF2) in different tumor cells. Methods Lung cancer cell line H1299 A549 and breast cancer cell line MCF-7 MDA-MB-435s were selected. The radiosensitivity index SF _ 2 of different cells was detected by cloning assay after irradiation of 2 Gy, the relative telomere length of different cells was measured by real-time fluorescence quantitative PCR, and 緯 -H2AX protein was labeled by immunofluorescence labeling method. The levels of endogenous DSBs damage in different cells were detected. The relationship between endogenous DSBs and radiosensitivity of tumor cells was evaluated by statistical analysis. Results Immunofluorescence labeling of 緯 -H2AX protein showed that there was no significant difference between H1299 and A549 in the endogenous DSBs level of four kinds of tumor cells except H1299 and A549 (P 0.05). There were significant differences between other two groups in the level of endogenous DSBs and SF _ 2. The correlation coefficients of telomere length were -0.95 and -0.97, respectively. Conclusion Endogenous DSBs of tumor cells can be used as a marker to predict radiosensitivity of tumor cells. The correlation between them may be related to the part of telomere involved in the production and repair of endogenous DSBs.
