輸注聚合人胎盤血紅蛋白對大鼠氧化損傷的初步研究

【摘要】:目的探討輸注不同濃度聚合人胎盤血紅蛋白(Poly PHb)對大鼠造成的氧化損傷。方法建立動物換血模型,將50只SD大鼠隨機均分為假手術組:僅做股動/靜脈插管處理;氯化鈉注射液(對照)組:股動/靜脈插管,輸注氯化鈉注射液行換血實驗;2%、4%及6%Poly PHb 3個實驗組:股動/靜脈插管,分別輸注相應濃度的Poly PHb行換血實驗。分別在大鼠換血前、換血70%后即刻觀測平均動脈壓(MAP),換血完成后即刻從大鼠股動脈取血0.5 m L/只,做動脈血血氣分析,并檢測各組大鼠血漿中還原型谷胱甘肽(GSH)濃度、超氧化物歧化酶(SOD)活力、總抗氧化能力(T-AOC)、丙二醛(MDA)濃度、8-羥基脫氧鳥苷(8-OHDG)濃度。結果 1)大鼠換血70%后,2%、4%及6%Poly PHb組與對照組相比,GSH(μmol/L)分別為33.76±14.33、47.98±21.88、39.73±15.54及10.25±5.70(P0.01);SOD(U/m L)分別為238.07±27.39、355.73±35.42、405.26±45.01及138.46±57.16(P0.01);T-AOC(U/m L)分別為20.50±7.87、32.65±4.38、38.03±4.50及5.40±2.22(P0.01)。2)大鼠換血70%后,2%、4%及6%Poly PHb組與對照組相比,MDA濃度(nmol/m L)隨著Poly PHb濃度的增高而增高:45.08±4.12、54.39±3.16、58.52±6.33及2.85±0.59(P0.01)。3)大鼠換血70%后,2%、4%及6%Poly PHb組與對照組相比,大鼠血漿8-OHDG濃度(nmol/m L)未發(fā)生明顯變化:2.54!0.90、2.76!0.99、2.79!0.82及2.26!0.42(P0.05)。結論 Poly PHb對大鼠造成脂質過氧化損傷,且氧化損傷程度隨著Poly PHb濃度的增高而加重,但并未對大鼠DNA造成損傷。
[Abstract]:Objective to investigate the oxidative damage induced by different concentrations of polymerized human placental hemoglobin (Poly PHb) in rats.Methods 50 SD rats were randomly divided into sham-operation group (femoral artery / vein intubation only) and sodium chloride injection (control) group (femoral / venous intubation).Three experimental groups: femoral artery / vein catheterization, transfusion of sodium chloride injection with 4% and 6%Poly PHb, and transfusion of corresponding concentration of Poly PHb were performed.The mean arterial pressure (MAPH) was measured immediately before and 70% after the exchange of blood in rats, and 0.5 mL of blood was taken from the femoral artery of rats immediately after the exchange of blood.
