帕瑞昔布鈉對膿毒癥小鼠腸黏膜屏障的保護作用

【摘要】:目的研究帕瑞昔布鈉對膿毒癥小鼠腸黏膜屏障功能的影響及可能機制。方法 21只C57BL/6小鼠隨機分為三組:假手術組(Sham組)、膿毒癥模型組(CLP組)、膿毒癥模型+帕瑞昔布鈉2mg/kg治療組(P組),每組7只。術后24h用襻環(huán)結扎法檢測各組小鼠小腸通透性。另21只C57BL/6小鼠隨機分為三組,分組及治療同前,術后24h處死小鼠,收集小鼠回腸。小腸組織行HE染色觀察腸病理損傷。采用Western bolt法檢測小腸ZO-1、Occludin、Claudin-1等緊密連接蛋白的表達。采用ELISA法檢測小腸黏膜IL-6、PGE2的濃度。結果與Sham組比較,CLP組小鼠小腸明顯損傷,門靜脈血內(nèi)FD4濃度明顯升高,小腸黏膜內(nèi)ZO-1、Occludin、Claudin-1蛋白表達明顯減少、IL-6與PGE2濃度明顯升高(P0.05)。與CLP組比較,P組小鼠小腸損傷明顯減輕(P0.05),門靜脈血內(nèi)FD4濃度明顯降低(P0.05),小腸黏膜內(nèi)各蛋白表達水平明顯增加、IL-6與PGE2濃度明顯降低(P0.05)。結論帕瑞昔布鈉治療可以明顯降低膿毒癥導致的腸黏膜屏障功能損傷,其機制可能與降低腸組織炎癥水平,增加腸緊密連接蛋白表達相關。
[Abstract]:Objective to study the effect of paroxib sodium on intestinal mucosal barrier function in septic mice and its possible mechanism.Methods Twenty-one C57BL/6 mice were randomly divided into three groups: sham group (sham group), sepsis model group (C57BL/6 group), paruoxib sodium 2mg/kg group (n = 7).The permeability of small intestine in each group was detected by loop ligation 24 hours after operation.The other 21 C57BL/6 mice were randomly divided into three groups. The mice were killed 24 hours after operation and the ileum was collected.The intestinal pathological injury was observed by HE staining.Western bolt method was used to detect the expression of cladin-1 and other tight junction proteins in the small intestine.The concentration of IL-6 and PGE2 in small intestinal mucosa was detected by ELISA method.Results compared with the Sham group, the small intestine injury and the FD4 concentration in the portal vein were significantly increased, and the expression of Claudin-1 protein in the intestinal mucosa was significantly decreased. The levels of IL-6 and PGE2 were significantly increased (P 0.05).Compared with CLP group, the injury of small intestine in P group was significantly alleviated, the concentration of FD4 in portal vein blood was significantly decreased, and the expression level of various proteins in intestinal mucosa was significantly increased. The levels of IL-6 and PGE2 in small intestine were significantly decreased (P 0.05).Conclusion Pareoxib sodium therapy can significantly reduce the intestinal mucosal barrier function damage induced by sepsis, and its mechanism may be related to the reduction of intestinal inflammation and the increase of intestinal tight junction protein expression.
